Objective:To study the effect of "Qingfei Xiegan Decoction" on lipid metabolism and the mechanism of pathological state of hyperlipidemia induced by high fat and high sugar in insulin resistance model rats with type 2 diabetes.Methods:60 five week old male SD rats were fed regularly and quantitatively in the animal feeding room of Yanbian University.60 rats were used for one week to adapt to the environment and maintain the appropriate temperature and humidity in the room.According to the body weight of rats,they were randomly divided into normal group,type 2 diabetes model group,metformin group,qingfeixiegan Decoction low dose group,middle dose group and high dose group,with 10 individuals in each group.Except for the normal group,50 rats were fed with high fat and high sugar diet.When the weight of rats increases slowly,the feed can be added.If necessary,high calorie nutritious foods such as melon seeds and walnuts can also be fed to help the weight increase.When the body weight of the rat reaches more than 400g,the next step of model preparation can be carried out.The rats were forbidden to feed and water on the night before modeling.The next day,streptozotocin(STZ)was injected intraperitoneally at a dose of 40 mg/kg to make the rats suffer from diabetes.72 hours later,it was also forbidden to provide feed the night before,but water was not allowed.Fasting blood glucose and body weight of rats were measured.If the blood glucose is not less than 11.1 mmol/L,the model can be established successfully.After the model was established successfully,gastric perfusion was started for 28 days,once a day.At the end of the gavage,the rats were forbidden to feed the feed for 12 hours,but could not help but water.They were anesthetized with chloral hydrate,took the supine position,shaved the chest,sterilized the skin,cut the "V" shaped abdomen with surgery,aimed at the heart,and injected the needle from the most obvious beating part of the heart.The injection depth was generally suitable for 2.5-3 mm.After blood collection,the serum was centrifuged.The contents of body weight,fasting blood glucose(FPG),triglyceride(TG),total cholesterol(TC),high-density lipoprotein cholesterol(HDL-C)and low-density lipoprotein cholesterol(LDL-C)in serum were measured,and the expression levels of insulin(INS),fatty acid(FFA)and leptin(LEP)in serum were determined by ELISA The degree of insulin resistance and dyslipidemia.Result:Compared with the normal group,there were significant changes in the contents of FPG,TG,TC and lp-c in the serum of STZ group.The results showed that the rats in the observation group had insulin resistance and had symptoms of hyperlipidemia and hyperglycemia,which made the insulin resistance of each observation group increased with the increase of leptin and free fatty acid content and the decrease of insulin content,indicating that the rats in the observation group had insulin resistance and the model was effective.According to the low,medium and high dose administration of chaoyao qingfeixiegan decoction,the serum FFA,TG,TC,LDL-C and HDL-C contents of metformin group and qingfeixiegan Decoction low,medium and high group were statistically different,and the values of each test index were reversed and improved to different degrees compared with the model group.Conclusion:"Qingfei Xiegan Decoction" can improve the disorder of lipid metabolism,reduce blood glucose and lipid,improve the insulin resistance of insulin resistance model rats,and play a certain role in the prevention and treatment of diabetes.
