Objective: Explore the effect of modified Jiawei Qingfei Xiegan Decoction on insulin resistance diabetic rats through nuclear factor-κB(NF-κB)signaling pathway,to provide a reference for the clinical treatment of insulin resistance diabetes.Methods: 50 clean healthy SD male rats were fed adaptively for one week.All the rats were randomly divided into normal group(n = 10)and model group(n = 40).The rats in the normal group were fed with normal diet.At the same time,the rats in the model group were fed with high-fat and high-sugar diet.Four weeks later,the insulin resistance model of type 2 diabetes was established after intraperitoneal injection of streptozotocin(STZ).40 rats were randomly divided into model group,low dose group of modified Qingfei Xiegan decoction and high dose group of modified Qingfei Xiegan decoction,metformin positive control group,followed by intragastric administration of corresponding drugs after the success of the model.The normal group and the model group were given the same amount of normal saline.The specimens were collected after 4 weeks of intragastric treatment.Blood samples were collected by abdominal aortic intubation under intraperitoneal anesthesia after 12 hours of fasting.After centrifugation was measured the high density lipoprotein(HDL-C),low density lipoprotein(LDL-C),total cholesterol(TC),triglyceride(TG).Moreover,the serum insulin index(INS)was detected by enzyme-linked immunosorbent assay kit.The tissue near the liver was isolated,and the liver tissue was used for the determination of interleukin-1β(IL-1β)and the Westernblot test of p65 protein and phosphorylated-IκBα(P-IκBα)protein after exposing the liver.Results: Compared with the model group,the Jiawei Qingfei Xiegan decoction group could reduce the serum insulin index and blood lipids of type 2 diabetic rats with insulin resistance to a certain extent(P <0.05).Moreover,it significantly reduced the liver tissue-associated inflammatory factor IL-1β and significantly decreased the protein expression of P65 protein and P-IκBα protein(P <0.05).Conclusion: Jiawei Qingfei Xiegan decoction can effectively improve the reduction of blood lipids and the protein expression levels of INS,IL-1β,P65 and P-IκBα in diabetic rats with insulin resistance.Therefore,Jiawei Qingfei Xiegan decoction can play a good role in the treatment and prevention of insulin resistance in type 2diabetes through NF-κB signal pathway.
