Objective:To evaluate the expression of Sox2 protein and HBXIP protein in upper tract urothelial carcinoma.Methods:The clinical data and paraffin specimens of 54 patients with upper tract urothelial carcinoma who were treated by surgery at the affiliated hospital of Yanbian university from January 2017 to September 2019 were collected.(16 of them had paraneoplastic urothelial tissue at the same time),10 patients with stricture of ureter underwent surgical treatment of normal upper urinary tract epithelium tissue paraffin specimens were be collected.The expression of Sox2 protein and HBXIP protein were detected by immunohistochemistry method in tissues of 54 upper tract urothelial carcinoma,16 paraneoplastic tissues and 10 normal upper urinary tract epithelium tissues.The relationship between levels of Sox2 protein and HBXIP protein expression and age,sex,clinical stage and pathological grade of UTUC patients was evaluated by chi-squared values.The correlation between levels of Sox2 protein and HBXIP protein expression in UTUC tissues was evaluated by spearman rank correlation test.54 patients after UTUC were followed up by phone call and outpatient reexamination.the shortest follow-up time was 4 months and the longest follow-up time was 36 months.Kaplan-Meier survival analysis and Log-Rank test were used to analyze the relationship between protein expression and survival time.Results:1.The positive expression rates of Sox2 protein in UTUC tissues and paraneoplastic urothelial tissues were 42.59%and 12.50%.No positive expression was found in normal upper urinary tract epithelium tissues.The difference of positive rate between UTUC tissue and paraneoplastic urothelial tissues was statistically significant(P<0.05).The positive expression rate of Sox2 protein in T2-T4 group was 57.14%,which was significantly higher than that in Ta-T1 group(26.92%);the comparison of the positive expression rate between the two groups was statistically significant(P<0.05).The positive expression rate of Sox2 protein in high-level group was 54.05%,which was significantly higher than that in low-level group(17.65%);the comparison of the positive expression rate between the two groups was statistically significant(P<0.05).Survival analysis showed that the median overall survival time of patients with positive expression of Sox2 protein was shorter than that of patients with negative expression of Sox2 protein(P<0.05).2.The positive expression rates of HBXIP protein in UTUC tissue,paraneoplastic urothelial tissue and normal upper urinary tract epithelium tissue were 79.63%,31.25%and 20.00%,The difference of positive rate between UTUC tissue and paraneoplastic urothelial tissues was statistically significant(P<0.05).The positive expression rate of HBXIP protein in T2-T4 group was 92.86%,which was significantly higher than that in Ta-T1 group(65.38%);the comparison of the positive expression rate between the two groups was statistically significant(P<0.05).The positive expression rate of HBXIP protein in high-level group was 89.19%,which was significantly higher than that in low-level group(58.82%);the comparison of the positive expression rate between the two groups was statistically significant(P<0.05).Survival analysis showed that the median overall survival of patients with positive expression of HBXIP protein was shorter than that of patients with negative expression of HBXIP protein(P<0.05).3.The positive expression level of Sox2 protein in UTUC tissue is positively correlated with the positive expression level of HBXIP protein.Conclusion:1.The positive expression of Sox2 protein and HBXIP protein is related to the occurrence and development of UTUC.2.The positive expression of Sox2 protein and HBXIP protein is related to the malignant degree of UTUC.3.The positive expression of Sox2 protein and HBXIP protein is related to the prognosis of patients with UTUC4.Detecting the expression rate of Sox2 protein and HBXIP protein in UTUC tissue may become one of the indicators of auxiliary diagnosis and prognosis judgment of UTUC.
