| OBJECTIVE: To investigate the distribution of CD36-rs1722505 SNPs in the Zhuang and Han populations in Guangxi Zhuang Autonomous Region(GZAR)and the correlation between the distribution of CD36-rs1722505 SNPs and coronary heart disease(CHD)and its risk factors.METHODS: The subjects of the study lived in Guangxi Zhuang Autonomous Region for a long time, and the parents of the zhuang people were all zhuang people, the parents of the han people were all han people, and there was no blood relationship between the individuals.Seventy-one patients who underwent coronary angiography in the department of cardiovascular medicine of our hospital from February 2019 to February 2020 were selected.According to the results of coronary angiography, the subjects of the study were divided into coronary artery smooth(control group)and coronary artery lesion(case group).According to the degree of coronary artery lesions, the people of case group were divided into one-vessel lesion,two-vessel lesion and three-vessel lesion. In the coronary artery disease group,58 patients(39 males and 19females)were aged(60.59±10.70)years.There were 19 cases of zhuang nationality(58.63±8.95 years old)and 39 cases of han nationality(61.54±11.45 years old). There were 17 patients in the one-vessel lesion(57.47±9.99 years old),22 patients in the two-vessel lesion(62.32±10.80 years old),and 19 patients in the three-vessel lesion(61.37±11.15 years old).In the smooth coronary artery group,13 patients(6 males and 7 females)were aged(62.54±7.77)years,6 were of zhuang nationality(61.17±10.30 years old),and 7 were of han nationality(63.71±5.38 years old). The genotype of CD36-rs1722505 SNPs was determined by PCR-RFLP.TC,TG,LDL-C,HDL-C,FBG,PBG,Hb A1 c and other laboratory indicators were measured by automatic biochemical analyzer. And the SPSS 21 statistical software was used for data statistical analysis.RESULTS: 1.A total of 71 patients who underwent coronary angiography(CAG)were enrolled. There were 25 zhuang people and the number of TT,TC and CC genotypes of cd36-rs1722505 was 1,15 and 9,accounting for 4.0%,60.0% and36.0%,and the allele frequency of T and C was 34.0% and 66.0%.The han nationality had 46 cases and the number of TT,TC and CC genotypes was 6,23 and 17,accounting for 13.0%,50.0% and 37.0%,and the allele frequency of T and C was 38.0% and 62.0%.There was no significant difference in genotype frequency and allele frequency between the two groups(P=0.395,P=0.633).2.There were 58 cases in the case group. The number of TT,TC and CC genotypes of cd36-rs1722505 was 6,31 and 21,accounting for 10.3%,53.5%and 36.2%,and the allele frequency of T and C was 34.6% and 65.4%.In the control group of 13 cases, the number of TT,TC and CC genotypes was 1,7 and5,accounting for 7.7%,53.8% and 38.5%,and the frequency of T and C alleles was 37.1% and 62.9%.There was no statistically significant difference in the frequency of each genotype and allele between the two groups(P=0.953,P=0.814).3.Compare different degree of coronary artery lesions,we found that the TC genotype accounted for the highest in the smooth coronary artery group,one-vessel lesion and two-vessel lesion, accounting for 53.8%,58.8%,59.1%.And among the three-lesions,the CC genotype accounted for the highest, accounting for 47.4%.The proportion of C allele in each group was higher than that of T allele. But the differences between genotype and allele were not statistically significant(P=0.935,P=0.839).The prevalence rates of hypertension and diabetes were significantly different among the groups(P<0.05).In the group with high degree of coronary artery disease, the prevalence of hypertension and diabetes were higher than those with smooth coronary artery disease and mild degree of coronary artery disease.4.Among the 17 patients with diabetes history, the number of TT,TC and CC genotypes was 0,10 and 7,accounting for 0.0%,58.8% and 41.2%,and the allele frequency of T and C was 29.4% and 70.6%.Among the 54 patients with no diabetes history, the number of TT,TC and CC genotypes was 7,28 and 19,accounting for 13.0%,51.8% and 35.2%,and the allele frequency of T and C was 38.9% and 61.1%.There was no statistical difference in the genotype frequency and allele frequency between the two groups(P=0.294,P=0.317).The prevalence of hypertension in people with a history of diabetes was significantly higher than that of people without a history of diabetes(P=0.047),and their systolic blood pressure was also significantly higher than that of people without a history of diabetes(P=0.035).5.We take the coronary artery lesions as dependent variable,at the same time to the age,sex(0,1),BMI,blood pressure(0,1),diabetes(0,1),smoking history(0,1),nationality(0-Zhuang,1-Han),systolic blood pressure, diastolic blood pressure, fasting glucose, glycosylated hemoglobin, total cholesterol, triglyceride, low density lipoprotein cholesterol,high-density lipoprotein cholesterol, the genotype of CD36-rs1722505(1-TT,2-CC,3-TC)as independent variables. And then we carried out Logistic stepwise regression analysis of the above variables. The results showed that none of the above covariates entered the equation.CONCLUSION: 1.The distribution of TT,TC,CC genotypes and T,C alleles of CD36-rs1722505 was basically the same in zhuang and han populations, patients with or without coronary artery disease or diabetes,as well as in patients with severity of coronary artery disease. However, there was no statistically significant difference in genotypes and alleles frequencies between the two groups. It is considered that this polymorphic locus of CD36 gene may not be the key factor affecting coronary artery disease and/or diabetes in zhuang and han populations of Guangxi.2.People with diabetes are more likely to develop hypertension than those without diabetes, and hypertension and/or diabetes are risk factors for increased coronary atherosclerosis. |
