Based On The Theory Of Oxidative Stress,to Explore The Effect Of Yishen Tongluo Recipe On Sperm DNA Integrity In Patients With Kidney Deficiency And Blood Stasis Infertility

Objective:To observe the clinical efficacy of Yishen Tongluo Recipe in the treatment of sperm DNA damage in patients with kidney deficiency and blood stasis infertility,and to explore its mechanism of action,to provide clinical data support for the development of Yishen Tongluo Recipe;to analyze sperm DNA fragment rate and semen parameters,ROS,PON-1 correlation.Methods:According to the order of treatment,80 patients who met the inclusion criteria were randomly divided into treatment group(Yishen Tongluo Recipe)and control group(Luocarnitine oral solution)by computer random number generator method.Among them,4 cases were excluded from the control group and treatment group,and 72 cases were included in the statistics.Analysis: 36 patients in the treatment group and the control group.The first day from the patient’s visit was recorded as the start date,and the 12-week session was a course of treatment.The treatment group was treated at the start date and after the end of the treatment period.Semen analysis parameters(average 2 times),seminal plasma ROS,PON-1 activity,sperm DFI were examined with the control group,and data were collected and statistical software was used for correlation analysis to explore the clinical efficacy of Yishen Tongluo Recipe.And the mechanism of action.Results:;(1)Correlation analysis: sperm DFI was negatively correlated with sperm survival rate,forward motion percentage and activity rate,and the correlation coefficients were-0.372,-0.341,-0.488(P <0.05).Sperm DFI and seminal plasma ROS were positive.Correlation coefficient was 0.556(P <0.05).Sperm DFI was negatively correlated with PON-1 activity,and the correlation coefficient was-0.467(P <0.05);(2)In the treatment group,36 cases were clinically cured,markedly effective,effective,and ineffective,respectively,5 cases,11 cases,18 cases,2 cases,the totaleffective rate was 94.44%;36 cases of the control group,the number of cured,markedly effective,effective,ineffective cases There were 3 cases,9 cases,12 cases and 12 cases,respectively.The total effective rate was 66.67%.The treatment group was superior to the control group by x2 test,and the difference was statistically significant(P <0.05).(3)After treatment,the sperm concentration,forward movement percentage,total vitality and other parameters of the two groups were improved,sperm DFI decreased significantly,and the treatment group was superior to the control group(P <0.05);(4)In terms of seminal plasma ROS and PON-1,both groups could reduce the ROS content of seminal plasma and increase the activity of PON-1.The treatment group was superior to the control group(P <0.05).Conclusion : Yishen Tongluo Recipe can significantly improve the sperm DNA integrity of patients with kidney deficiency and blood stasis type;Yishen Tongluo Recipe can enhance PON-1 enzyme activity,improve the body’s antioxidant capacity,reduce the ROS content of seminal plasma,significantly reduce sperm DFI,improve semen Analysis of concentration,forward motion,total vitality and other parameters.
