ObjectiveBy observing the mitochondrial morphology and enzymes in the liver tissues of kidney-deficiency rats induced by gavage thyroxine tablets and reserpine suspension by observing the Zuogui pill group and its split group(ziyin group and buyang group)The effect of activity,to explore the correlation between the syndrome of kidney-yin deficiency and energy metabolism,and to reveal the mechanism of Zuogui pill“Yangzhongqiuyin”in the treatment of kidney-yin deficiency syndrome,to study Zuogui pill and its split group,Buyang group)To explore the scientific connotation of“seeking yin in yang”on the changes of mitochondrial structure and enzyme activity in liver tissue of kidney-yin deficiency rats.MethodsFifty six SPF male SD rats were selected for one week adaptive feeding in the laboratory.Except for the control group,the remaining rats were given thyroxine tablets and reserpine by gavage to prepare kidney yin deficiency model.The experimental animals were randomly divided into model group,Ziyin group,Buyang group and Zuogui pill group.Ziyin group,Buyang group and Zuogui pill group were given by gavage.Zuogui pill group was given the crude drug dose of 9.45g/kg,the Ziyin group was given the crude drug dose of 7.29g/kg,and the Buyang group was gavaged with crude drug dose of 2.16g/kg,and the volume of gavage was 7.5m L/kg.The control group and model group were given equal volume normal saline once a day.After successful intervention,blood samples were collected from inferior vena cava and liver tissue lines were extracted The activities of Na~+-K~+-ATPase,LDH,SDH,CCO and ATP content in liver mitochondria were measured by ELISA.The changes of mitochondrial structure in liver tissue were observed under electron microscope,and the ratio of camp to c GMP in serum was determined.Results1 Changes in the ratio of c AMP to c GMP in serum of rats in model group:compared with the control group,the ratio of c AMP to c GMP in serum of rats in model group increased significantly(P<0.01).2 Effects of zuogui pill and its decoction group(ziyin group and buyang group)on the enzyme activity and ATP content of liver mitochondria in rats with kidney-yin deficiency syndrome:Compared with control group,model group rats liver mitochondrial enzyme Na~+-K~+-ATPase,SDH and LDH,CCO activity and ATP content were increased,the significant difference(P<0.01),compared with model group,Yin and Yang group,left to pill group liver mitochondrial enzyme Na~+-K~+-ATPase,LDH,SDH and CCO activity and ATP content decreased,statistically significant difference(P<0.05 or P<0.01),and left to pill group curative effect best(P<0.01).3 Changes in the mitochondrial structure of liver tissue in experimental rats of each group:Compared with the control group,the mitochondria of the model group were significantly swollen,the ridge arrangement was disordered,and the rupture disappeared;compared with the model group,the pathological damage of the rat liver mitochondria was significantly reduced,And the Zuogui Pill group had the best effect,close to the control group.Conclusion1 The mixed suspension method of gavage thyroxine tablets and reserpine can successfully prepare the kidney yin deficiency syndrome model.2 After drug intervention in the model group,the mitochondrial Na~+-K~+-A-TPase,SDH,CCO activity and ATP content in liver tissue decreased,suggesting that the Zuogui pill group and its split group may act on mitochondria by r-egulating enzyme activity To improve the body’s energy metabolism level,and then play a therapeutic role.3 Zuogui pill and its split group may affect the mitochondrial energy met abolism level by improving the mitochondrial pathological structure.Therefore,the change of mitochondrial structure is also one of the targets that affect the energy metabolism level.4 The Zuoguiwan Quanfang group is superior to the simple disintegrating Ziyin group and the disintegrating Buyang group in treating kidney-yin deficie-ncy syndrome,confirming the compatibility of Zuoguiwan’s“seeking yin in yang”and further embodying The law’s compatibility value.
