Objective:To observe the changes of hepatic mitochondrial energy metabolism induced by hydrocortisone in kidney Yang deficiency rats,to explore the improvement of hepatic mitochondrial energy metabolism in kidney Yang deficiency rats by jinhuihuishengqi pill and its disposer,and to study the mechanism of its action to treat kidney Yang deficiency,to provide modern scientific basis for the basic theory of " Junior fire supporting vital qi " in traditional Chinese medicine,and to to reveal the essence of Junior fire supporting vital qi " from the perspective of system biology,to reveal the law of differentiation and treatment,and also to provide a more scientific experimental basis for the extensive application of shenqi pillMethods:A total of 60 SD rats(male and female)were fed with common feed,48 of which were treated with hydrocortisone intraperitoneal injection and 2weeks preparation of kidney Yang deficiency syndrome.According to the random grouping principle,the rats could be divided into model group,kidney Qi group,kidney Qi Pill qu Fugui group and Fugui group.Add 12 blank groups.Among them,the kidney gas pill group,the kidney gas pill to Fugui group,Fuzi Guizhi group with 7.29 m L/Kg gavage 1 day,continuous 2weeks;Normal group and model group were given7.29 m L/Kg,once a day for 2 weeks.On the first day of the model,all rat body weights were measured on the first day and on the fourteenth day of the gavage,anal temperature was measured on the fourteenth day of the gavage,all rats were anaesthetized and killed on the morning after the end of the gavage,liver tissue was isolated,ATP,ADP and AMP were measured by HPLC,the mitochondrial structure of liver tissue was observed by electron microscopy,and enzyme immunoassay(ELISA)was used to determine liver mitochondria.The contents of ATP,ADP and AMP in liver mitochondria of rats were determined by HPLC,and the activities of hepatic mitochondrial respiratory complex Ⅰ-Ⅳ-ATP,PPH,LDH,PK,HK,ICCT,ICD,CS,ACS,alpha-KGDHC were determined by ELISA.Result:(1)After the success of the model,the rats appeared chills Yang deficiency performance in the symptoms and signs: fear of cold,back curl,hi pile,hair shedding,perianal pollution,reduced intake of food and drinking water,delayed movement,and reduced activity.(2)Compared with normal group,the content of ATP and ADP in PFH,LDH,PK,HK,ICD,CS,α-KGDHC,respiratory chain complex Ⅰ-Ⅳ activity and liver mitochondria decreased significantly and the difference was statistically significant(P < 0.05)Compared with the model group,the prognosis of kidney gas pill increased significantly,and the difference was statistically significant(P < 0.05).Compared with the model group,there was no statistically significant difference between the groups of shengqi pill qu Fugui and Fugui(P(P >0.05).(3)Compared with the normal group,the mitochondrial structure of the model group was incomplete,even had cavitation denaturation,some mitochondrial edema,the disorder of the crest arrangement,even the disappearance of the crest,the electron density became lower,the matrix color was lighter,compared with the model group,the ultrastructure of the liver mitochondrial structure was obviously improved,manifested in the decrease of mitochondrial edema,the majority of mitochondrial structure was normal,the groups of shengqi pill qu Fugui and Fugui mitochondrial structure disorder,appear vacuole-like degeneration.Conclusions:(1)The content of ATP and ADP in liver mitochondria of rats with kidney Yang deficiency decreased,and there were mitochondrial energy metabolic disorders.(2)The levels of ATP and ADP in the respiratory chain complex Ⅰ-Ⅳ activity and liver mitochondrial were recovered,the content of AMP decreased and the EC increased after the intervention of kidney-gas pills.The pathological changes of the ultrastructure of liver mitochondria were improved.Thus regulating the mitochondrial function and energy metabolism of rat liver,which may be the modern biological basis of the combination of " Junior fire supporting vital qi " of kidney-gas pills to treat kidney-Yang deficiency syndrome,thus providing scientific experimental basis for the treatment of kidney-Yang deficiency syndrome by traditional Chinese medicine.
