Objective:To study the effect of Cassia Twig and Dragon Bone Combination on blood lipid and Hcy in rats with autonomic nerve function impairment and carotid atherosclerosis,and to further observe the clinical efficacy of this decoction,so as to provide new ideas for the treatment of patients with autonomic nerve function impairment and.carotid atherosclerosis.Methods:This study is divided into two parts:animal experiment and clinical research.In animal experiments,the rats were given adaptive feeding for 1 week.After 1 week,blood samples of orbital inner canthus were collected to detectblood lipid.A total of 60 SPF grade male SD rats(200-240g)were randomly divided into the following 5 groups:control group,model group,lippert group,sham operation group,and cassia twig and keel oyster soup treatment group,with 12 rats in each group.Local autonomic nerve damage was caused bydetachment of the cervical sympathetic nerve trunk.In addition to the control group and the sham operation group,the other three groups were treated with cervical sympathetic nerve trunk dissection immediately after 1 week of adaptive feeding to establish a rat model of autonomic nerve dysfunction.The sham group was anesthetized only,and the nerves were stripped but not ligated.Atherosclerosis model establishment:high-fat diet was fed for 12 weeks,15g~20g/day,and vitamin D3 was intramuscular injected at a total dose of 700000IU/kg at week 1,3 and 7 of the first feeding.Modeling success:thickening of arterial intima or lipid striations were found as the success indicators of modeling.Each group was fed high-fat diet for 12 weeks on the second day after the operation of cervical sympathetic nerve trunk dissection.Aftersuccessful modeling,the group was given cassia twig and keel oyster soup by gavage of 10.7g/kg and lml/100g once a day for 6 consecutive weeks.In the lipitor group,the dose of 6mg/(kg·d)and 1ml/100g were given by gavage once a day for 6 weeks.The control group,model group and sham operation group were given the same amount of normal saline,once a day for 6 weeks.Serum was collected at week 12 and 18 respectively.The levels of LDL,HDL,trigl yceride,total cholesterol and Hcy were measured with automatic biochemical instrument.The level of potential change before and after modeling was monitored.In the clinical part,60 patients from Qingdao haici medical group with carotid atherosclerosis and autonomic nerve dysfunction were randomly divided into treatment group and control group,30 cases each.Treatment group patients were treated with atorvastat in calcium tablet,Cas sia Twig and Dragon Bone Combination at the same time by Qingdao city hospital of traditional Chinese medicine TCM pharmacy unified allocate free Fried particles after packaging.One bag at a time,add 200mL warm water,and rinse after breakfast and dinner.Patients in the control group received atorvastatin calcium tablets(20mg/tab let,Pfizer pharmaceutical co,LTD.)for 20mg/time and 1 time/day,and both groups were treated for half a month.The changes of scopa-aut scores before and after treatment were observed.Changes of LDL and homocysteine in the two groups before and after treatmentResults:In the experimental part,lipid profile:(1)at the end of the 12th week,the total cholesterol,triglycerides and LDL in the model group,the sham operation group,thelipi tor group and the Cassia Twig and Dragon Bone Combination group were all higher than the control group,with statistical sign ificance(P<0.05).(2)at the end of the 18th week,the total cholesterol,triglyceride and low-density lipoprotein in the lipitor group,the Cassi a Twig and Dragon Bone Combination group were all lower than those in the modelgroup and the sham operation group,with statistically significant differences(P<0.05).In addition,the triglyceride,LDL and total cholesterol levels of rats in the Cassia Twigand Dragon Bone Combination group at the 18th week were alllower than those at the 12th week,which had statistical significance(P<0.05).At week 18,the level of HDL in the Cassia Twig and Dragon Bone Combination group was significantlyhigher than that in the model group and the sham group,withstatistically significant difference(P<0.05).(3)serum Hcylevel:compared with the model group,the Hcy level of the lipitor group and the Cassia Twig and Dragon Bone Combination group were significantly lower at the end of the 18th week,with statistically significant differences(P<0.05).Moreover,the Hcy level in the treatment group at week 18 was lower than that in the treatment group at week 12,showing statistical significance(P<0.05).In the cl inical part,the score of autonomic nerve function scale significantly decreased in the treatment group after treatment,while the score showed no significant change in the control group after treatment.After treatment,the score of autonomic nerve function scale in the treatment group was lower than that in the control group,with statistical difference(P<0.05).The serumHcy level changed significantly after treatment compared with the control group,and the difference before and after treatment was significant in the treatment group(P<0.05).There was a significant difference in the level of LDL after the treatment(P<0.05),and there was a significant difference in the level of LDL in the control group before and after thetreatment(P<0.05).Conclusions:(1)in clinical practice,the scopa-aut scale score of patients with autonomic nerve dysfunction combined with carotid atherosclerosis was significantly reduced in the Cassia Twig and Dragon Bone Combination group,and the levels of low-density lipoprotein and homocysteine were alsodecrea sed.(2)Cassia Twig and Dragon Bone Combination and lipitor can significantly reduce the levels of TC,TG,LDL and hcy in rats,and Cassia Twig and Dragon Bone Combination can improve the level of HDL,so it is believed that the use of the two drugs together may have a better role in regulating blood lipid. |
