Objective:From the perspective of traditional Chinese medical theory and clinical practice,it is considered that kidney yang deficiency is of great significance in the evolution of pulmonary fibrosis pathogenesis,and the therapeutic effect of Kidney Yang warming combined with Tonifying Qi,eliminating phlegm,promoting blood circulation and dredging collaterals is considerable.In the study of pathology and pathogenesis of pulmonary fibrosis in modern medicine,it is found that abnormal angiogenesis may accelerate the progress of pulmonary fibrosis.VEGF,as an angiogenic factor,can interfere with the formation of pulmonary fibrosis.The previous experiment of the research group confirmed that icariin can inhibit the expression of VEGF,angiogenesis and slow down the development of pulmonary fibrosis.In this study,we observed the effect of Icariin on the proliferation of pulmonary microvascular endothelial cells in vitro,explored the effect and mechanism of Icariin on angiogenesis,observed the gene and protein expression of key factors of VEGF related pathway,and verified that icariin,a traditional Chinese medicine,plays an anti pulmonary fibrosis role by intervening angiogenesis and searching for the target of action,so as to provide experimental basis for clinical medication.Through the analysis of typical cases,to explore the clinical treatment of pulmonary fibrosis patients with kidney yang deficiency and the mechanism,and summarize the clinical experience of tutors.In order to provide new ideas and experimental basis for the treatment of pulmonary fibrosis,theoretical research,clinical analysis and experimental research are combined.Methods:CCK-8 method was used to detect the effect of Icariin on the proliferation of pulmonary microvascular endothelial cells and its maximum non-toxic concentration.Transwell cell chemotaxis experiment and scratch healing experiment were used to study the effect of Icariin on the migration of pulmonary microvascular endothelial cells.ELISA method was used to detect the concentration of VEGF in the supernatant of cells to detect the intervention of Icariin on the secretion of VEGF The expression of VEGF gene was detected by time PCR,and the expression of VEGF,PLC γ 1,PI3K and Akt were detected by Western blot.Results and Conclusion:Icariin can inhibit the proliferation and migration of pulmonary microvascular endothelial cells,reduce the expression level of angiogenesis related factors VEGF,PLC γ1,PI3K,Akt.It is speculated that icariin may inhibit the angiogenesis through the intervention of VEGF and other related signal pathways,so as to slow down the progress of pulmonary fibrosis,and provide experimental theoretical basis for the prevention and treatment of pulmonary fibrosis by Icariin.
