Effects Of Acupotomy On The Expression Of IL-1β,TNF-α And ASIC1 In A Rabbit Model Of Scapulohumeral Periarthritis

Periarthritis of shoulder is the abbreviation of periarthritis of shoulder,which is called "off shoulder wind" and "shoulder congealing disease" in traditional Chinese medicine.Modern medicine also calls it "frozen shoulder" and "adhesive arthritis".It is a common and frequently occurring disease in the middle-aged and old people.Periarthritis of shoulder is a chronic aseptic inflammation caused by local soft tissue degeneration and strain.Its clinical manifestations are shoulder pain with limited activity.Muscle adhesion and atrophy can be seen in the elderly.ObjectivesThe study of continuous mechanical strain add ice compress method preparation of periarthritis of shoulder the rabbit model with needle knife overall solutions for operative intervention,and,as compared with the blank group and model group rabbit observation pine needle knife method for periarthritis of shoulder,tissue morphology of rabbit model of local behavior,and pathological changes and joint synovial IL-1 beta,TNF alpha and ASIC1,the influence of the level of gene and protein expression to explore the possible mechanisms of needle knife therapy for the periarthritis of shoulder intervention effect,provide experimental basis for treatment of needle knife release the disease.MethodsThirty male New Zealand rabbits were randomly divided into blank group,model group and acupotomology group,with 10 rabbits in each group.A rabbit model of periarthritis of shoulder was established by continuous mechanical strain and ice compress.After successful modeling,rabbits in the acupotomology group were treated with acupotomology overall release intervention for 4 weeks.After the end,the expression,posture,hair and shoulder joint activity of New Zealand rabbits in each group were observed,and the pathological changes of shoulder joint tissue were observed by HE staining section.The gene and protein expression levels of interleukin(IL)-1β,tumor necrosis factor(TNF)-α and acid-sensing ion channel 1(ASIC1)in shoulder synovium of each group were detected by RT-PCR and Western blot,respectively.Results Acupotomology release method can improve the local pathological state,reduce the synovium and muscle interstitial cellulose exudation,adhesion and mechanization.Compared with blank group,the expression levels of IL-1β,TNF-α and ASIC1 gene and protein in synovial membrane of shoulder joint in model group were significantly increased(P <0.0001).Compared with model group,the expression levels of IL-1β,TNF-α and ASIC1 gene and protein in synovium of shoulder joint of rabbit in acumotomology group were significantly decreased(P <0.0001).ConclusionsAcupolomology may reduce the gene and protein expression levels of IL-1β,TNF-α and ASIC1 in the synovium of shoulder joint by improving the pathological changes of local tissue,reduce the inflammation and pain sensitivity of local tissue,and play a therapeutic role in the treatment of periarthritis of shoulder by anti-inflammatory and analgesia.Due to the greater degree of motion of the shoulder joint,the muscle involved in the activity is more,as well as the shoulder joint itself anatomical characteristics and biomechanical reasons,so the rehabilitation of the shoulder joint disease is more difficult than other joints,such as the use of a single treatment method,it is difficult to achieve the ideal effect.Small needle knife therapy small needle knife therapy is the "needle" acupuncture therapy and surgery of "dao","dao" can directly release the main point of soft tissue adhesions,reduce inflammation regional organizations internal pressure,blocking its vicious stimulating of nerves,blood vessels,relieve or correct state of hypoxia and ischemia,improving local blood fluid circulation,to restore the disorder of dynamic balance of local organizations,plus the stimulation of "needle" dredge the meridian,qi and ventilation,balance of Yin and Yang,the general principles of the indolence,radically cure lesions.Extrapolation is the traditional reduction technique.It is mainly used to repair fracture and dislocation.The author will improve the method of extubation and extension for the treatment of periarthritis of shoulder by multi-directional extubation and extension technique.Combined with static and static,dissipate blood stasis and detumination,invigorate blood and relax tendons,the general principle is painless and shoulder arbi can be cured.
