| Objective: To study the role of NLRP3 inflammatory corpuscles and their downstream cytokines in the pathogenesis of psoriasis complicated with MS,and to explore the relationship between psoriasis and MS pathogenesis.Methods: 22 patients with psoriasis vulgaris,22 patients with MS in dermatology and venereal department,22 MS patients in endocrine department or hypertension department and 22 healthy control patients were collected.The patients and healthy volunteers were given 10 ml of EDTA anticoagulant and5 ml of anticoagulant respectively,and the treatment and detection were carried out respectively.The specific methods were as follows: 1.EDTA anticoagulant cryocentrifugation,the supernatant plasma was frozen at-80℃,and PBMC was separated by lymphocyte separation solution in the lower layer;the blood serum was frozen at-80℃without anticoagulant centrifugation.2.Trizol method was used to extract the total RNA from PBMC,and the expression of ASC,caspase-1 and NLRP3 m RNA in inflammatory corpuscles was detected by RT-q PCR.3.the expression of IL-1 β,IL-17,IL-18 and tnf-αin the serum of the above patients and control groups were detected by ELISA.Results: 1.There was no significant difference in gender and age among psoriasis group,psoriasis combined with MS group,MS group and control group(P > 0.05);there was no significant difference in PASI score between psoriasis group and psoriasis combined with MS group(P > 0.05).2.RT q PCR results:(1)ASC in PBMC There was no interaction between psoriasis and MS.(2)the expression of Caspase-1 m RNA in peripheral blood mononuclear cells in psoriasis group was higher than that in control group(P < 0.05).There was no significant difference between psoriasis and MS group and control group(P > 0.05)There was no significant difference between MS group and control group(P > 0.05).There was no interaction between MS group and psoriasis.3.ELISA results: the serum levels of IL-1β,IL-17,IL-18 and TNF-α in the case group were higher than those in the control group(P < 0.05);the serum levels of IL-1β,IL-17 and IL-18 in the case group were as follows:MS group > psoriasis combined with MS group > psoriasis group;the serum levels of TNF-α in the case group were as follows: psoriasis combined with MS group > MS group >psoriasis group;there was interaction between psoriasis and MS,which jointly affected the serum levels of IL-1β The secretion of IL-17,IL-18 and TNF-α.4.Correlation analysis results: the levels of IL-1β,IL-17 and IL-18 in serum were positively correlated with caspase-1 and NLRP3 m RNA in PBMC,and negatively correlated with ASC m RNA.There was no significant correlation between TNF-α and ASC,caspase-1,NLRP3 m RNA.Conclusion: 1.NLRP3 and its downstream cytokines IL-1β,IL-18 and IL-17 are related to the pathogenesis of psoriasis with Ms.it is speculated that NLRP3 inflammasome may play a pro-inflammatory role by mediating its downstream inflammatory cytokines IL-1β,IL-18 and IL-17.2.TNF-α participates in the pathogenesis of psoriasis with MS through non NLRP3 inflammasome pathway.3.Psoriasis and MS interact in promoting the secretion of IL-1β,IL-17,IL-18 and TNF-α. |
