Effect Of IL-4 On Proliferation,Invasion And Migration Of Gastric Cancer SGC7901 Cells Via STAT6 Pathway And Its Mechanism

ObjectiveTo explore the effect of IL-4 on proliferation,invasion and migration of gastric cancer SGC7901 cells and its possible mechanism.MethodsGastric cancer SGC7901 cells subcultured in vitro,and divided into control group,IL-4 group,IL-4+AS1517499 group,and AS1517499 group.The final concentration of exogenous IL-4 was 5,10,20,30,40 ng/ml,respectively.The final concentration of STAT6 specific inhibitor AS1517499 is 1.2μmol/L.After 48 hours of starvation culture,MTT assay was used to detect cell proliferation ability,cell scratch assay was used to detect cell migration and adhesion ability,Transwell invasion assay was used to detect cell invasion ability,and Western blotting was used to detect STAT6 total protein and phosphorylated STAT6(p-STAT6).Expression,Western blotting and RT-q PCR were used to detect the protein and m RNA expression of MMP-7,CD34 and E-cadherin.Results1.Compared with the control group,after the addition of exogenous IL-4,the cell proliferation,migration,scratch healing,and invasion ability of the IL-4group was significantly increased(P<0.01),and the cell proliferation ability of the IL-4+AS1517499 group and the AS1517499 group,Invasion ability and scratch healing ability were lower than IL-4 group(P<0.01).2.Western blotting experiments showed that compared with the control group,the expression of p-STAT6,MMP-7 and CD34 protein in the IL-4 group increased,and the expression of E-cadherin protein decreased(P<0.01),compared with the IL-4 group,in the IL-4+AS1517499 group,the expression of p-STAT6,MMP-7 and CD34 protein decreased,and the expression of E-cadherin protein increased(P<0.01).There was no significant difference in the total protein expression of STAT6(P>0.05).3.q PCR experiments showed that,compared with the control group,the expression of MMP-7 and CD34 at the m RNA level increased in the IL-4 group,and the expression of E-cadherin at the m RNA level decreased(P<0.01),compared with the IL-4 group,in the IL-4+AS1517499 group,the expression of MMP-7 and CD34 at the m RNA level decreased,and the expression of E-cadherin at the m RNA level increased(P<0.01);the STAT6 pathway specific inhibitor AS1517499 can inhibit the above-mentioned effects of IL-4.ConclusionsIL-4 may increase the expression of MMP-7 and CD34 protein through the STAT6 pathway and reduce the expression of E-cadherin protein,thereby promoting the proliferation,invasion and migration of gastric cancer SGC7901 cells.
