Objective:To investigate the expression changes of several important molecular markers in breast cancer tissues before and after neoadjuvant chemotherapy and the effects of these changes on disease-free survival and 5-year survival rate of patients. Methods:A total of 822 samples of patients diagnosed with breast cancer for the first time were collected. All the collected data were from patients treated in the Affiliated Cancer Hospital of Xinjiang Medical University during 2013-2019,and the treatment method was neoadjuvant chemotherapy followed by radical surgery.specimens were patients diagnosed before and for the first time for ultrasound guided biopsy specimens, specimens after chemotherapy(and once again the specimen)for complete tumor specimens, postoperative routine immunohistochemical examination on the patients and the prognosis of patients with full including data collection, line comparison before and after neoadjuvant chemotherapy patients prognosis and expression of molecular markers of change, judge whether there is some influence or contact between them. Results: The data showed that neoadjuvant chemotherapy did affect the expression of some molecular markers, among which the presentation of ER,HER-2 and Ki-67 molecular markers crucially changed, with statistical significance(P<0.05),and the expression level of PR and Ki-67 was statistically significant(P<0.05);Furthermore,the expression alter of ER,PR and Ki-67 reflected in the life expectancy of patients, among which the indicator Ki-67 expression had the most obvious effect on the prognosis of patients, and the decrease of its expression indicated a good prognosis of patients.Although MP grade suggested that neoadjuvant chemotherapy had no obvious effect about the overall survival and survival time(OS)of patients(P>0.05),but had a momentous effect on tumor-free survival, with significantly prolonged DFS(P<0.05).Conclusion:Changes in ER and PR expression can reflect the prognosis of patients to a certain extent, while Ki-67 index data reflect the data changes and prognosis of patients with excellent performance.Several biomarkers indicated that neoadjuvant chemotherapy could significantly prolong the time with tumor progression survival of patients.Several important molecular biomarkers mentioned in the data could reflect the sensitivity of patients to chemotherapy drugs and treatment methods, but their roles and mechanisms still need further study.Neoadjuvant chemotherapy is newer and less studied than conventional adjuvant therapy, but the deeper potential of this newly emerging therapy has not been fully discovered. With the continuous exploration of the direction of clinical trials, neoadjuvant chemotherapy can better guide treatment in clinical practice, and has a broad therapeutic prospect in new directions such as helping patients screen available drugs.
