| Objective:To observe the pathological changes of lung tissue and the protein expression levels of IKK α,I κB a and p65 in nuclear transcription factor(NF-κb)signaling pathway in mice infected with RSV,and to explore the mechanism and dose effect relationship of flavonoids in Qingfei oral liquid in the treatment of RSV pneumonia in mice.Methods:Fifty BALB/c mice were randomly divided into five groups:normal group,model group,high-dose flavone group,low-dose flavone group and ribavirin group.The normal group was given physiological salt water,the other four groups were infected with RSV by nasal drip,the virus concentration was 10-4/50ul.Then the drug groups were prepared with high-dose flavone group 120mg/kg/d,low-dose flavone group 40mg/kg/d,ribavirin group 50mg/kg/d,lasting for 4 days.The mice were sacrificed on the fourth day after treatment.The lung tissue sections were taken to observe the inflammatory changes.The serum IL-6 content in the lung tissue was detected by enzyme-linked immunosorbent assay(ELISA).The expression levels of NF-κB IKK α,I κB α and p65 were determined by polymerase chain reaction(PCR),immunohistochemistry(IHC)and Western blot(WB).Results:Compared with the normal group,NF-κB in the model group decreased significantly The protein expression of IKK α,IκBα and p65 increased significantly(P<0.05),and the mRNA trend increased(P<0.05),which was consistent with the characteristics of inflammatory reaction;compared with the model group,the serum IL-6 level decreased in the high-dose and low-dose flavone groups and Western medicine ribavirin groups(P<0.05),and the index protein level decreased in each group(P<0.05),and there was no significant concentration gradient difference between the high-dose and low-dose flavone groups.Conclusion:Compared with ribavirin,the flavonoids in Qingfei oral liquid have similar or better efficacy,which may be related to the intervention of the disease process after the flavonoids in Qingfei oral liquid enter the body,By affecting the regulation of NF-κB signaling pathway and reducing the expression of NF-κB protein,it can effectively reduce the synthesis and release of cytokines,improve the inflammatory reaction in the lung tissue of mice,and achieve the therapeutic effect of reducing the structural and functional damage of lung tissue. |
