| Objective 1.To observe the division and differentiation of bone marrow mesenchymal stem cells(BMSCs)in rat thin uterus.2.To observe and compare the homing of bone marrow mesenchymal stem cells assisted by simple injection and electroacupuncture.Methods 40 SD rats were divided into four groups:control group,model group,cell group and acu+cell group.With the exception of the control group,95%ethanol intrauterine injection method was used to make the model.Cell suspension injection through caudal vein was performed in the cell group and the acu+cell group on the day of modeling,the third day after surgery,the second estrus phase,and the third estrus phase.The dosage is 1*106/ml,1 ml per time.In the acu+cell group,except for cell injection,electroacupuncture was performed daily on Guanyuan,Zigong and Sanyinjiao for ten consecutive days.The wave form was selected as density wave.The stimulation intensity was 1mA,the frequency was 2/15Hz,and the time was 15min.During this period,the changes of body weight and estrus phase were continuously recorded in each group.Uterine specimens were taken after three oestrous cycles,and uterine coefficient was calculated.Endometrial thickness,number of blood vessels and number of glands were measured by HE staining.Cell proliferation index Ki67 was detected by immunohistochemistry,and the expression of estrogen receptor and progesterone receptor was detected by Western-blot.RT-PCR was used to quantitatively analyze the expression of SRY gene in rat endometrium.Results The expression of SRY gene in the combined group was significantly higher than that in the cell group,and the difference was statistically significant.Compared with the model group,the endometrial thickness of the cell group and the combined group was significantly thicker,and the number of vessels and glands on the endometrium were significantly increased,with statistical significance.There was no significant difference in estrus cycle recovery among the four groups except the blank group.Compared with model group,uterine coefficient in cell group and combined group was significantly increased.The expression level of Ki67 in cell group and combination group was significantly higher than that in model group.The expression of estrogen receptor in cell group and combined group was significantly higher than that in model group.The expression level of progesterone receptor in cell group and combined group was significantly higher than that in model group,and the expression level of progesterone receptor in combined group was significantly higher than that in cell group.Conclusion Electroacupuncture can promote the homing of stem cells,electroacupuncture combined with bone marrow mesenchymal stem cell injection can promote the proliferation of rat thin endometrium cells,endometrium thickening,endometrial blood vessels,glands and estrogen progesterone receptor recovery. |
