| ObjectiveBased on the method of network pharmacology,the biological network of Modified Buzhong Yiqi Decoction(BZYQTJJ)in the treatment of Allergic Rhinitis(AR)was constructed and analyzed,and the active ingredients and potential targets of Modified Buzhong Yiqi Decoction for application to ARwere discussed.MethodsBased on pharmacokinetic parameters(ADME)and supplemented by literature,the chemical constituents of Modified Buzhong Yiqi Decoction were screened by TCMSP database.Using TCMSP/PubChem/SwissTargetPrediction/Uniprot platform to retrieve potential targets for chemical composition,through the OMIM/DRUGBANK/Genecard/TDD/DisGeNET database screening get AR related targets.The String database was used to extend the protein network related to AR and construct the PPI network.The Cytoscape software was used to build the relationship network of "BZYQTJJ-active ingredients-targetsdisease" and the topology of the network was analyzed to identify the effective components and core targets of Modified Buzhong Yiqi Decoction in the treatment of AR.Autodock Vina was used to conduct molecular docking,and the binding activity of key chemical components to core targets was verified.David database was used to carry out GO function enrichment analysis and KEGG biological pathway enrichment analysis on the targets of Modified Buzhong Yiqi Decoction in the treatment of AR,so as to further explore the mechanism of action of Modified Buzhong Yiqi Decoction in the treatment of AR.ResultsThe Modified Buzhong Yiqi Decoction has a therapeutic effect on AR by acting on the core targets of IL-6,TNF,CXCL8,VEGFA,PTGS2,IL-1B,IL-10,CCl2,TLR4,IL-4 and other active components such as quercetin,kaempferol and luteolin etc.According to P<0.05,FDR<0.05 and the sequencing of gene counts,screeningthe top 20 G0 functional items,speculated that Modified Buzhong Yiqi Decoction plays a significant role in the treatment of AR by participating in inflammatory response,positive regulation of transcription from RNA polymerase Ⅱ promoter,signal transduction,response to drug and other biological processes,combining with proteins binding,enzymes binding and identical protein binding.According to P<0.05,FDR<0.05 and the sequencing of gene counts,the top 20 KEGG pathways were screened,and it was speculated that Modified Buzhong Yiqi Decoction could treat AR by regulating multiple signaling pathways,such as TNF signal pathway,PI3K-Akt signal pathway and Toll-like receptor signal pathway etc.ConclusionThis study preliminarily explored the effective components and mechanism of Modified Buzhong Yiqi Decoction for application to AR based on the molecular level,which is expected to provide new ideas and basis for the subsequent research and promote the further development of traditional Chinese medicine in the treatment of Allergic Rhinitis. |
