Effect Of Zhu Lian Slow Twisting Acupuncture On The Expression Of SOD,CAT And Apoptosis In Liver And Kidney Tissues Of Aging Model Rats

Objective:To observe the effect of Zhu Lian slow twisting acupuncture on peroxidation and apoptosis of liver and kidney tissues in aging model rats,and to explore its mechanism of delaying aging.Methods:60 male SD rats were randomly divided into blank control group,model control group,Zhu Lian slow twisting acupuncture prevention group,common acupuncture prevention group,Zhu Lian slow twisting acupuncture treatment group,and common acupuncture treatment group using random number table,with 10 rats in each group.Except for the blank control group,the other five groups of rats were intraperitoneally injected with D-galactose to establish a subacute aging model.At the same time of modeling,the prevention group was treated with Zhu Lian slow twisting acupuncture and common acupuncture for 42 consecutive days.At the end of modeling,the treatment group was treated with acupuncture using the corresponding method for 28 consecutive days.The content of H₂O₂in rat liver and kidney tissues was detected by colorimetry;the content of MDA in rat liver and kidney tissues was detected by thiobarbituric acid assay（TBA）;the mRNA expression of SOD and CAT in rat liver and kidney tissues was detected by quantitative real-time PCR（q-PCR）;the expression of SOD and CAT protein in rat liver and kidney tissues was detected by Western blot（WB）;and the apoptosis rate of liver and kidney tissues was detected by Terminal-deoxynucleoitidyl Transferase Mediated Nick End Labeiling（TUNEL）.Result:（1）Compared with the blank control group,the contents of H₂O₂and MDA in the liver and kidney of rats in the model control group were significantly increased（P<0.05）;Compared with the model control group,the contents of H₂O₂and MDA in the liver and kidney of rats in the prevention groups and treatment groups were significantly decreased（P<0.05）.There is a statistically significant difference between the two prevention groups,the H₂O₂and MDA contents of the Zhu Lian slow twisting acupuncture group were lower;There is a statistically significant difference between the two treatment groups,the H₂O₂and MDA contents of the Zhu Lian slow twisting acupuncture group were lower;There is a statistically significant difference between the two Zhu Lian slow twisting acupuncture groups,the H₂O₂and MDA contents of the prevention group were lower;There is a statistically significant difference between the two common acupuncture groups,the H₂O₂and MDA contents of the prevention group were lower（P<0.05）.（2）Compared with the blank control group,the mRNA and protein expressions of SOD and CAT in the liver and kidney of rats in the model control group were significantly decreased（P<0.05）;Compared with the model control group,the mRNA and protein expressions of SOD and CAT in the liver and kidney of rats in the prevention groups and treatment groups were significantly increased（P<0.05）.There is a statistically significant difference between the two prevention groups,the mRNA and protein expressions of SOD and CAT of the Zhu Lian slow twisting acupuncture group were higher;There is a statistically significant difference between the two treatment groups,the mRNA and protein expressions of SOD and CAT of the Zhu Lian slow twisting acupuncture group were higher;There is a statistically significant difference between the two Zhu Lian slow twisting acupuncture groups,the mRNA and protein expressions of SOD and CAT of the prevention group were higher;There is a statistically significant difference between the two common acupuncture groups,the mRNA and protein expressions of SOD and CAT of the prevention group were higher（P<0.05）.（3）Compared with the blank control group,the apoptosis rate of liver and kidney tissues in the model control group was significantly increased（P<0.05）;Compared with the model control group,the apoptosis rate of liver and kidney tissues in the prevention groups and treatment groups was significantly reduced（P<0.05）.There was no significant difference in liver apoptosis rate between the two prevention groups and between the two treatment groups（P>0.05）;There is a statistically significant difference in apoptosis rate of kidney tissues between the two prevention groups,the apoptosis rate of kidney tissues of the Zhu Lian slow twisting acupuncture group was lower;There is a statistically significant difference between the two treatment groups,the apoptosis rate of kidney tissues of the Zhu Lian slow twisting acupuncture group was lower（P<0.05）.There is a statistically significant difference between the two Zhu Lian slow twisting acupuncture groups,the apoptosis rate of liver and kidney tissues in the prevention group was lower;There is a statistically significant difference between the two common acupuncture groups,the apoptosis rate of liver and kidney tissues in the prevention group was lower（P<0.05）.The apoptosis rate of liver tissues in the same group was lower than the kidney tissues’（P<0.05）.Conclusions:（1）Both Zhu Lian slow twisting acupuncture and common acupuncture can reduce the contents of H₂O₂and MDA in liver and kidney tissues of aging rats.（2）Both Zhu Lian slow twisting acupuncture and common acupuncture can increase the gene and protein expressions of SOD and CAT in liver and kidney tissues of aging rats.（3）Both Zhu Lian slow twisting acupuncture and common acupuncture can reduce the apoptosis rate of liver and kidney tissues in aging rats.（4）Zhu Lian slow twisting acupuncture and common acupuncture have obvious preventive and therapeutic effects on aging rats,and the preventive and therapeutic effects of Zhu Lian slow twisting acupuncture on aging rats are superior to those of common acupuncture in some aspects.（5）The mechanism of Zhu Lian slow twisting acupuncture in delaying aging may be related to the resistance to tissues peroxidation,thereby reducing apoptosis.
