In recent years,phototherapy mainly includes photothermal therapy and photodynamic therapy.As a new type of anti-bacterial and anti-tumor therapy,it has been widely concerned.It is not only a special treatment,but also can solve the problem of drug resistance caused by traditional drugs,and has the advantages of small side effects and no damage to normal cells.This paper mainly uses gold nanorods with good light-to-heat conversion efficiency as the matrix substrate.The gold nanorods are coated with silica layer and amination modified,and the photosensitizer is loaded on the gold nanorods through electrostatic adsorption/chemical coupling.Obtained nanomaterials with PDT-PTT combined therapeutic effect.The following is the main work of this paper:(1)PDT/PTT based on NIR mediated gold nanorods loaded with Rose Bengal(RB)Using gold nanorods as a carrier,a layer of loose and porous dielectric material SiO2is coated on the surface of the gold nanorods and modified by amination,and then the photosensitizer Rose Bengal(RB)is loaded to obtain the nanomaterial AuNR@SiO2-NH2-RB.Photothermal and singlet oxygen performance test,antibacterial test,drug uptake imaging,hemolysis rate test and normal cell dark toxicity test were carried out to characterize AuNR@SiO2-NH2-RB.The results showed that:AuNR@SiO2-NH2-RB had good photothermal effect and singlet oxygen capacity.The variation temperature(ΔT)of AuNR@SiO2-NH2-RB can reach 42℃,and the final temperature can exceed 74℃,whileΔT and the final temperature of AuNR@SiO2-NH2are 32℃and 62℃,respectively.The order of singlet oxygen release capacity was:RB>AuNR@SiO2-NH2-RB>AuNR@SiO2-NH2.This may be due to the fact that part of RB has been wrapped in the AuNR@SiO2-NH2with lots of pores.Under the illumination,the concentration of 5μM of AuNR@SiO2-NH2-RB showed a inhibition rate of 95.12%towards Escherichia coli,which can kill 98.20%of Staphylococcus aureus at 1μM.For both of Gram-negative bacteria and Gram-positive bacteria,the antibacterial ability order was:AuNR@SiO2-NH2-RB>RB>AuNR@SiO2-NH2.The results showed that AuNR@SiO2-NH2-RB possessed good PDT/PTT effect.(2)PDT/PTT of gold nanorods loaded with chlorin e6(Ce6)based on NIRUsing gold nanorods as a carrier,a layer of loose and porous dielectric material SiO2is coated on the surface of the gold nanorods and modified by amination.Then the photosensitizer chlorin e6(Ce6)is loaded to obtain the nanomaterial AuNR@SiO2-NH2-Ce6.Photothermal and singlet oxygen performance test,antibacterial test,drug uptake imaging,hemolysis rate test and normal cell dark toxicity test were carried out to characterize AuNR@SiO2-NH2-Ce6.The results showed that:AuNR@SiO2-NH2-Ce6 had good photothermal effect and singlet oxygen energy.The change temperature(ΔT)can reach 40℃,and the final temperature can exceed 72℃,whileΔT and final temperature of AuNR@SiO2-NH2were 32℃and 62℃,respectively.The singlet oxygen release capacity order rate of 97.56%against Escherichia coli and 1μM of AuNR@SiO2-NH2-Ce6 can kill 99.96%of Staphylococcus aureus.For both Gram-negative bacteria and Gram-positive bacteria,the order of antibaterial ability was:AuNR@SiO2-NH2-Ce6>Ce6>AuNR@SiO2-NH2.The resultes showed that AuNR@SiO2-NH2-Ce6 exhibited good PDT/PTT effect.(3)PDT/PTT of AuNR@SiO2-NH2-Ce6-VES based on NIRUsing gold nanorods as a carrier,a layer of loose and porous dielectric material SiO2is coated on the surface of the gold nanorods and modified by amination.Then the vitamin E succinic acid(VES)and the photosensitizer chlorin e6(Ce6)are loaded to obtain the nanomaterial AuNR@SiO2-NH2-Ce6-VES.Photothermal and singlet oxygen performance test,cell acticity test were carried out to characterize AuNR@SiO2-NH2-Ce6-VES.The results showed that:AuNR@SiO2-NH2-Ce6 had good photothermal effect and singlet oxygen energy.The change temperature(ΔT)can reach 40℃,and the final temperature can exceed 70℃,whileΔT and final temperature of AuNR@SiO2-NH2were 32℃and 62℃,respectively.The singlet oxygen release capacity order was:Ce6>AuNR@SiO2-NH2-Ce6-VES>AuNR@SiO2-NH2.This may be due to the fact that part of Ce6 has been wrapped in the pores of AuNR@SiO2-NH2.After treatment with AuNR@SiO2-NH2-Ce6-VES,photodynamic therapy,photothermal therapy and combination therapy were carried out AuNR@SiO2-NH2-Ce6-VES,and results of CCK-8 method showed that the inhibition rate of SMMC-7721 cells increased with the increase of concentration of AuNR@SiO2-NH2-Ce6-VES.When the concentration of AuNR@SiO2-NH2-Ce6-VES was 8μM,the inhibition rate under PDT combined with PTT condition was 80.38%,which was higher than that under PDT alone(46.47%)or PTT alone(74.53%).The results of FCM showed that the level of ROS in SMMC-7721 cells increased after treatment AuNR@SiO2-NH2-Ce6-VES The level of ROS in SMMC-7721 cells was 79.4%at the concentration of 8μM,indicating its photodynamic effect.The resultes showed that AuNR@SiO2-NH2-Ce6-VES exhibited good PDT/PTT effect.
