| Objective To establish a rat model of intrauterine adhesion by mechanical curettage,evaluate the effectiveness of the model,and study the therapeutic effect of platelet-rich plasma on intrauterine adhesion in rats.Methods Female SD rats were used as the experimental research object.Through mechanical curettage established the rat model of intrauterine adhesion.Set up four groups: normal saline group,blank control group,platelet-rich plasma group,sodium hyaluronate group,with9 rats in each group.The intrauterine adhesion of uterine specimens was observed by naked eye observation,the number of endometrial glands was detected by HE staining,and the degree of endometrial fibrosis was detected by Masson staining.The contents of TGF-β1 and MMP-9 in tissues were detected by ELISA,and the effect of platelet-rich plasma on the prevention of uterine adhesion in rats was detected at animal level.Results 1.General view of uterine tissue: The uterine surface of the control group was smooth,ruddy,uniform in thickness and good elasticity,and there was no adhesive band in the uterine cavity,which was easy to separate.In the model group,the uterine surface was rough,the color was white,the thickness was uneven,and the elasticity was poor.Adhesion bands were found in the uterine cavity,which were difficult to separate.The performance of platelet rich plasma group and sodium hyaluronate group was between the normal group and the normal saline group,and the difference between the two groups was not obvious.2.The number of endometrial glands was measured by HE staining:compared with the control group(8.11±4.07/HP),the number of endometrial glands in the model group(1.89±1.91/HP)was lower,(P<0.05).Compared with the model group,the number of glands in the platelet-rich plasma group(8.93±4.84/HP)was more(P<0.05).Compared with the sodium hyaluronate group(10.26±5.00/HP),the number of glands in the platelet rich plasma group was lower(P <0.05).3.The area of endometrial fibrosis detected by Masson staining:Compared with the control group(63.61±5.50%),the area of endometrial fibrosis in the model group(75.70±4.61%)was larger(P <0.05).Compared with the model group,the area of fibrosis in the platelet rich plasma group(68.18±6.49%)was less(P <0.05).Compared with sodium hyaluronate group(54.83±8.89%),the area of fibrosis in platelet rich plasma group was larger(P <0.05).4.TGF-β1 content in uterine tissue detected by ELISA: Compared with the control group(14.19±14.51pg/ml),TGF-β1 content in model group(79.93±57.34 pg/ml)was higher(P<0.05).Compared with model group,the content of TGF-β1 in platelet-rich plasma group(91.16±57.34pg/ml)was higher(P>0.05).Compared with sodium hyaluronate group(53.62±45.07pg/ml),the content of TGF-β1 in platelet rich plasma group was higher(P>0.05).5.ELISA detection in uterine tissue of MMP-9 content in tissues:Compared with the control group(272.84±160.51pg/ml),the content of MMP-9 in model group(117.91±71.79pg/ml)was lower(P<0.05).Compared with model group,the content of MMP-9 in platelet-rich plasma group(214.33±89.36 pg/ml)was higher(P<0.05).Compared with sodium hyaluronate group(129.71±36.12pg/ml),the content of MMP-9in platelet rich plasma group was higher(P>0.05).Conclusion 1.The rat model of uterine adhesion can be established by mechanical curettage.2.Platelet-rich plasma can prevent the formation of uterine adhesion in rats. |
