| Objective: To study the effects of different concentrations of Lycium barbarum polysaccharide(LBP)on the proliferation and expression of vascular endothelial growthfactor(VEGF)in human retinal vascular endothelial cells(HRCEC)under high glucoseenvironment in vitro.Methods:After enucleation of eyeball,HRCEC were selected and cultured under high glucose conditions in vitro,and Logarithmic growth cells were selected for the following experiments.Experimental groups: 1g/L low glucose group,4.5g/L high glucose group,4.5g/L high glucose and 20μg/m L LBP group,4.5g/L high glucose and 40μg/m L LBP group,4.5g/L high glucose and 80μg/m L LBP group.These five groups were cultured for 12,24 and 36 hours respectively.After different concentrations of LBP treatment in high glucose environment for 12,24 and 36 hours,CCK-8 method was used to detect the proliferation activity of HRCEC.The expression of VEGF in HRCEC was detected by Western Blot.Results:(1)Proliferation activity of HRCEC was detected by CCK-8,and detected the expression of VEGF by Western Blot.These results showed: At the same culture time,the optical density value of HRCEC and the expression of VEGF in the higher glucose group were remarkably higher than those in the lower glucose group(P<0.05);At same culture time,the optical density value of HRCEC and the expression of VEGF in the intervention groups with different concentrations LBP(20μg/ml LBP,40μg/ml LBP,and 80μg/ml LBP)were significantly lower than those in the high glucose group,suggesting that LBP had inhibitory effect on the proliferation of HRCEC,and the effect was stronger in the LBP concentration80μg/ml group than the LBP 20μg/ml group and LBP 40μg/ml group;In the same LBP concentration,after different action time,it was shown that the inhibition of LBP to HRCEC after 36 hours was stronger than that of 12 hours and 24 hours.(2)The Western Blot results are as follows: HRCEC was incubated in vitro environment for12 h,24h,and 36 h,comparing the gray value of VEGF,the expression of VEGF in the high glucose group was higher than that in the low glucose group(P<0.05);Under the same culture time,comparing the gray value of VEGF,the expression of VEGF in HRCEC of high glucose and different concentrations of LBP(20μg/ml,40μg/ml,80μg/ml)group was lower than that of high glucose group(P<0.05);Under the same incubation time,the high sugar and different concentrations of LBP plus drug group were compared in pairs;Comparing the gray value of VEGF,when the LBP concentration was80μg/ml,the expression of HRCEC VEGF was higher than that of the LBP 20μg/ml group and the LBP 40μg/ml group Lower(P<0.05);In the same concentration of LBP group at different times,comparing the gray value of VEGF,the expression of VEGF in HRCEC after 36 hours of intervention was lower than that in 12 hours and 24 hours(P<0.05).Conclusion:High concentration glucose environment,compared with small concentration glucose environment,is more favorable to the proliferation of HRCEC and up-regulate VEGF expression;In high glucose environment,different concentrations of LBP can inhibit the proliferation of HRCEC and down-regulate the expression of VEGF,and positively correlated with action time. |
