Objective Liver cancer is a common malignant tumor in our country, where mortality rate is high. It is a great threaten to residents, but there is no effective treatment for liver cancer. Lycium is one kinds of wolfberry shrub plants, which contains an active substances called LBP. Preliminary studies suggested that LBP can inhibit tumor growth. The purpose of this study is to explore the inhibition of LBP to growth of liver cancer cells,and expression of VEGF protein, which is a essential factor for the growth of tumor blood vessels.It is the hope to find some potential drugs for liver cancer treatment.Methods1. The high metastatic cell line HCCLM3 were cultured in DMEM medium.HCCLM3 cells were divided into control groups and LBP treatment groups(5umol/L,10umol/L,15umol/L),the cell proliferation was measured by MTT after 24 hours.2. The expression of VEGF protein was detected by Westernbolt after 24 hours. 3. HCCLM3 cells were divided into control groups and LBP treatment group(0umol/L,20umol/L),The expression of VEGF protein was detected by Immunofluorescence.Results1. In HCCLM3 cells,when the concentration of LBP is 0umol/L, 5umol/L,10umol/L,15umol/L . the inhibition rate is 0.80%,20.06%,35.44%,55.39%.(P<0.01)the conclusion is the cell proliferation was inhibited by LBP.2. The expression of VEGF protein can be apparently inhibited by LBP.With the increase of LBP concentration,the expression of VEGF protein increased gradually and in a dose depende nt manner.3. When the concentration of LBP reach at 20umol/L, the expression of VEGF protein can be completely inhibited by LBP,.ConclusionsLBP seems to inhibit the growth of liver cancer and expression of VEGF in vitro.
