| Objective: To observe the effect of jiedu prescription on the expression of Notch1,Hath-1 and MUC2 protein in mice with ulcerative colitis,and to investigate the mechanism of jiedu prescription in mice with ulcerative colitis.Methods: This experiment selected 40 male c57bl/6 mice,randomly selected 8 as blank group(n),and the rest of the mice established a uc model by freely drinking dss solution.the remaining model-making mice were randomly divided into three groups after the successful modeling was determined: model group(m),mesalazine group(s),and xuezhiedu prescription group(z),each group of 8.with the exception of the n group,each group was treated with corresponding drug gavage.after 14 d,all mice were sacrificed.The pathological changes of colon tissues were observed by hematoxylin-eosin(he)staining,the expression of muc2 protein in colon tissues was observed by immunohistochemistry,and the expression of notch1 and hath-1 protein in intestinal epithelial cells was detected by Western blotting(wester blot).Result:1.The general condition:Compared with the mice in group N,the mice in group m had mental state burnout,mucous pus in stool.After 14 days of treatment,the mental state of mice in group M was not improved.2.Disease activity index(DAI)score in miceCompared with the n(0.69±0.27)group,the DAI scores of mice in each group were significantly increased and the difference was significant(p <0.05).after 14 days of treatment,compared with the M group(6.08±5.18),the DAI scores of the rats in each treatment group were significantly reduced,and the difference was significant(p <0.05).Compared with s group(9.75±0.61),there was no significant difference in DAI scores in Z group(9.37±0.98)mice(p >0.05)3.colon mucosal index(CMDI)scoreCompared with the N group(0.16±0.35),the CMDI scores were significantly increased in each group(p <0.05).after treatment,compared with the M group(2.50±0.93),the CMDI scores of the other treated groups were significantly reduced and the difference was significant(p <0.05).there was no significant difference in CMDI scores between mice in S group(1.50±0.53)and Z group(1.38±0.52)(p >0.05).4.colon tissue he score in miceCompared with the N group(0.25±0.46),the colonic he scores wer e significantly increased in each group(p <0.05).after treatment,compa red with the M group(2.75±0.70),the colonic he scores of the other tr eatment groups were significantly reduced(p <0.05).There was no signi ficant difference in colonic he scores between S group(1.13±0.64)and Z group(1.00±0.53)(p >0.05)5.Effect of jiedu prescription on MUC2 protein in miceCompared with the blank group,the expression of MUC2 in colonic epithelial tissue of the model group was significantly weakened,and the distribution area of positive cells was significantly decreased(p <0.05).after administration,compared with the model group,the expression of MUC2 in both the positive control group and the xiezhuo jiedu prescription group was enhanced,and the distribution range of positive cells was increased(p <0.05).there was no significant difference between the positive control group and the xiezhuo jiedu prescription group(p >0.05).6.Effect of Xiezhujiedu recipe on mouse Notch1 and Hath1 proteinCompared with the blank group,the expression of Notch1 protein in the model group increased significantly,and the protein of Hath-1 decreased significantly(p <0.05).Compared with the model group,the expression of Notch1 protein in the positive control group and the Xiehujiedu group decreased significantly,and the protein of Hath-1 increased significantly(p<0.05).Conclusion:1.The expression of Hath-1 and MUC2 in colon epithelial cells d ecreased,and the expression of Notch proten increased.2.The general condition of UC mice was improved,the mucosal and pathological scores of colon were decreased,the expression of Hath-1 and MUC2 in colon epithelial cells was increased,and the expression of Notch1 protein was decreased,which indicated that it was effective for UC treatment3.The mechanism of Xiehujiedu prescription for the treatment of UC may be to protect the intestinal mucosal barrier by regulating the expression of Hath-1 and MUC2 protein in colon epithelial cells,reducing the expression of Notch1 protein and then improving the intestinal mucosal barrier... |
