| Objectives:Gastric Cancer(GC)is one of the most common gastrointestinal malignancies.Due to the absence of typical symptoms and effective molecular biomarkers,most patients with gastric cancer cannot be diagnosed early.Advanced endoscopic technology and medical management have improved the survival rate and prognosis of patients with early gastric cancer,but the death rate of patients with late gastric cancer is still high,and the 5-year overall survival rate is still low.Carcinogenic effect and progression of gastric cancer is a complex progress.Currently,the underlying mechanisms of gastric cancer development are to fully understand.Investigate the molecular mechanisms and signaling pathways related to gastric cancer may be contribute to identify potential diagnostic biomarkers and therapeutic targets.This paper aims to detect the expression level of target circRNA in gastric cancer tissues and gastric cancer cells,investigate the relationship between its expression level and the clinicopathological characteristics of gastric cancer,clarify its value in clinical diagnosis,so as to further study the biofunctional effects of target circRNA on cell cycle,proliferation,migration and invasion were further investigated.Methods:1.The expression differences of hsa_circ_0067934 in 70 pairs of gastric cancer tissues and their para-cancer tissues were detected through quantitative Reverse TranscriptionPolymerase Chain Reaction(q RT-PCR)technology.Information of clinicopathological features of 70 patients was recorded,and the correlation between the expression of hsa_circ_0067934 in gastric cancer tissues and the clinicopathological characteristic information was analyzed to build ROC(receiver operating characteristic curve)and study the clinical diagnosis value.2.The expression level of hsa_circ_0067934 in normal gastric mucosal epithelial cell lines(GES-1)and four gastric cancer cell lines(BGC-823,HGC-27,AGS,SGC-7901)was detected through q RT-PCR technology,and the gastric cancer cells was transfected transiently in vitro through small interfering RNA(si RNA)to study its biological function.3.The effect of down-regulating hsa_circ_0067934 on cell proliferation was detected by CCK-8 experiment;the effect of down-regulating hsa_circ_0067934 on cell cycle was detected by flow cytometric analysis;the effect of down-regulating hsa_circ_0067934 on cell migration was detected by scratch test and Transwell cell migration experiment;the effect of down-regulating hsa_circ_0067934 on cell invasion was detected by Transwell cell invasion experiment.Results:1.The expression level of hsa_circ_0067934 in gastric cancer tissues of 70 patients with gastric cancer was significantly higher than that in para-cancer tissues.Combined with the clinicopathological characteristics of the patients,the expression level of hsa_circ_0067934was closely related to the size of gastric cancer tissues,lymph node metastasis,and TNM stage,and was not related to age,gender,or degree of differentiation.By constructing the ROC curve,the area under the curve of hsa_circ_0067934 was 0.668,with clinical diagnosis value.2.The expression levels of hsa_circ_0067934 in four types of gastric cancer cells(bgc-823 hgc-27 AGS sgc-7901)were significantly higher than that of GES-1.It is in conformity with organizational outcomes,among which the expression difference in sgc-7901 was the most significant.3.The results of CCK-8 cell proliferation experiment found that cell proliferation was significantly inhibited by down-regulation of hsa_circ_0067934;the results of flow cytometry indicated that down-regulation of hsa_circ_0067934 affects cell cycle distribution,blocking cells in G1 phase;the results of scratch test and Transwell experiment of cell migration showed that down-regulation of hsa_circ_0067934 inhibits cells’ ability to migrate;and the Transwell experiment of cell invasion revealed that the ability of cell to invade was significantly inhibited after down-regulating hsa_circ_0067934.Conclusion:1.Hsa_circ_0067934 is highly expressed in gastric cancer tissue,which has certain value in clinical diagnosis and may be served as an effective biomarker.2.From the results of cellular functional experiments,it is indicated that down-regulating hsa_circ_0067934 plays an effective role in inhibition of proliferation,invasion and migration of gastric cancer cells,preventing it transitioning from G1 phase to S phase.3.Hsa_circ_0067934 is of great significance in occurrence and development of gastric cancer,being expected to a new tumor marker and therapeutic target in the future. |
