| The source of the plant is the dried roots of Peucedanum praeruptorum Dunn.It is mainly produced in Zhejiang,Henan,Jiangxi,Hunan,Sichuan and other places.P.praeruptorum Dunn tastes bitter,spicy and slightly cold."Medical Doctor’s Record"is classified as Chinese medicine,which has the functions of evacuating wind-heat,reducing qi and reducing phlegm.It is mostly used as a medicine component of trachea and bronchitis of wind-heat,cough,phlegm and asthma,and is one of the commonly used Chinese medicines.P.praeruptorum Dunn has more than 1500 years of medicinal history in China.Scholars have conducted a lot of research on it,especially in terms of chemical composition and pharmacological effects,and found that the main active ingredient of Qianhu is coumarin.It has anti-myocardial ischemic effect,vasodilator,blood pressure lowering,expectorant and other pharmacological effects.The anti-myocardial ischemia and antihypertensive effects of Praeruptorin A,B,C,and E were compared.It was found that all four coumarins had vasodilator effects.Praeruptorin C had the strongest effect and showed broad pharmacological aspects.The P.praeruptorum used in this experiment is Jiangxi Shangrao Xin-Qianhu.Shangrao was called Xinzhou in the past.Shangrao produce P.praeruptorum Dunn was also called"Xin-Qianhu"because of its thick roots,softness,black skin,yellow meat,and strong aroma.It is a traditional herbal medicine in Jiangxi.Radix Peucedani is rich in medicinal materials,and its pharmacological effects are extensive.With the advantages of modern instruments and data analysis technology in identifying chemical components and separation,Qianhu medicinal materials can be better developed and find the difference between the Xin-Qianhu and other Qianhu.So we have carried out a systematic chemical composition study on P.praeruptorum.Roots of Xin-Qianhu(13 kg)was extracted with Et OH repeatedly by impregnation method.These extracts were decompressed and concentrated,and then dissolved in water.The solvent was successively extracted with petroleum ether,trichloromethane,ethyl acetate and n-butanol,1210 g of petroleum ether,61 g of chloroform,25 g of ethyl acetate,and 135 g of n-butanol were obtained.Compounds were isolated by silica gel column chromatography,ODS column chromatography,Sephadex LH-20 column chromatography and semi preparative HPLC.The structures of compounds were identified by MS,NMR analysis(1H and 13C-NMR)and spectroscopic data.In order to further clarify the material basis of P.praeruptorum’s medicinal effect,it will provide a certain theoretical basis for the further development and utilization of Jiangxi Xin-Qianhu.32 compounds were isolated from ethanol extract of P.praeruptorum,which were23 pyrancoumarins:(±)-cis-4’-ethyl-3’-tigloylkhellactone(1),(3S,4S)-3-angeloyloxy-4- hydroxy-3,4-dihydroseselin(2),hyuganin B(3),selinidin(4),(±)-praeruptorin A(5),qianhucoumarin J(6),praeruptorin E(7),(±)-praeruptorin B(8),hyuganin C(9),corymbocoumarin(10),hyuganin D(11),pteryxin(12),qianhucoumarin I(13),qianhucoumarin D(14),(9R,10R)-9-acetoxy-8,8-dimethyl-9,10-dihydro-2H,8H-benzo[1,2-b:3,4-b′]dipyran-2-one-10-yl-ester(15),(+)-samidin(16),(±)-peuformosin(17),isoepo xypteryxin(18),(±)cis-4’-acetyl-3’-crotonoykhellactone(19),(3’S,4’S)-3’-O-isobutyroyl-4’-O-isovaleroylkhellactone(20),cis-3’,4’-diisovalerylkhellactone(21),qianhucoumarin C(22),(+)-ciskhellactone(23);1 linear pyrancoumarin compound:decursinol angelate(24);4furanocoumarins:deltoin(25),praeroside I(26),bergaten(27),xanthotoxin(28);1 simple coumarin:osthenol(29);3 othercompounds:[4-[β-D-Apiofuranosyl-(1→6)-β-D-glucopyr anosyloxy]-3-methoxypropiophenone](30),3-(4’-formylphenoxy)-4-methoxybenzald ehyde(31),bis(2-ethylhexyl)phthalate(32).Compounds 1-3,9,18-20,25,29-32 were isolated from this plant for the first time.It further enriched the chemical composition of P.praeruptorum and prepared the foundation for subsequent pharmacological screening. |
