Objective:To observe the rescue effect of hupehensis on Aconitum poisoning in vivo and in vitro,in order to provide scientific basis for rational utilization of hupehensis and provide methods for prevention and treatment of Aconitum poisoning.Method:1.To observe the effect of hupehensis on acute toxicity of Aconitum:determine the LD 50 of water extract of Aconitum,give the water extract of hupehensis in groups,and observe the poisoning behavior and death rate by grouping.2.To explore the effect of hupehensis on the cardiotoxicity of Aconitum poisoning:To observe the effect of hupehensis on the electrocardiogram of rats with acute Aconitum poisoning.H9c2 rat cardiac myocytes were cultured,the effects of hupehensis on aconitine induced myocardial cells were observed,the content of aconitine in cell fluid was detected,and the increment inhibitory effect of hupehensis on H9c2 cells of Aconitum was determined by MTT.Changes of lactate dehydrogenase and myocardial enzymes were measured by microplate method.Result:1.The LD 50=62.10mg/kg of water extract of Aconitum and mg/kg(600 mg/kg)could significantly reduce the toxic effect of Aconitum extract(60mg/kg).In advance,the water extract of hupehensis(water extract)could reduce the mortality from 50%to 20%,and the blood concentration of aconitine was reduced to 30.2%.2.The water extract of hupehensis can significantly improve the arrhythmia of rats,reverse the change of electrocardiogram of Q and ST-T and improve the arrhythmia.There is no significant difference in the content of aconitine in each cell liquid,and the retention rate is about 0.02mg/m L;the inhibition rate of 400 aconitine g/ml on H9c2 cells is 30.78%,and the content of lactate dehydrogenase is 12.50 U/L.The water extract of hupehensis can obviously damage the cardiac muscle cells.The inhibition rates of low,medium and high dose groups of the Chinese flowering crabapple were reduced to 24.61%,15.88%and 15.28%respectively,and the contents of lactate dehydrogenase were 8.73 U/L,7.63 U/L,7.56 U/L.respectively.In vivo experiments showed that the water extract of hupehensis could significantly reduce the toxicity of Aconitum on the internal muscles.After the Aconitum poisoning,the creatine kinase activity in mice increased to 2.44 U/m L,and the creatine kinase in the low,medium and high dose groups of Begonia decreased to 1.82U/m L,1.73U/m L and 1.59U/m L.respectively.Conclusion:1.The water extract of hupehensis can relieve Aconitum poisoning.The water extract of hupehensis can reduce the blood concentration of aconitine in the Aconitum and protect the target organs of Aconitum poisoning.It can be used to control Aconitum poisoning.2.The water extract of hupehensis can protect the myocardium.The water extract of hupehensis can reduce the damage of aconitine to myocardial cells and improve the symptoms such as arrhythmia.Its role is related to anti-oxidative damage and inhibition of lipid peroxidation,and can be used for cardiovascular protection..
