Nearly 20 million people are newly diagnosed with cancer every year,and the number of people who are still alive within five years of being diagnosed with cancer is only half of the number of people diagnosed with cancer.Among many types of cancer,oral cancer is a disease that is easily neglected by people.One of the early symptoms of oral cancer is oral ulcer,and most patients with oral cancer will ignore this symptom,resulting in the delay of the disease.Therefore,early diagnosis of oral cancer and its differentiation from conventional oral ulcers is now the top priority in oral cancer diagnosis.For the early in vitro diagnosis of oral cancer,there are CTC(circulating tumor cells),ct DNA(circulating tumor DNA),exosomes and other detection methods.However,for the simultaneous detection of oral cancer and oral ulcer,exosomes are the best choice.In contrast,several common exosome detection methods,NTA(Nanoparticle Tracking Technology)data acquisition is difficult and the process is complex;FCM(flow cytometry)detection of particle diameter is limited and cannot include all the range of exosome sizes.In general,all the existing detection methods have certain limitations,which is not conducive to the promotion of the practical application of exosome detection.In this paper,for the detection of exosomes in oral cancer and oral ulcer,a microfluidic chip that can detect multiple exosomes at the same time is proposed to be designed,and the injection automation of microfluidic chip is realized in combination with professional expertise in the process of experimental development.This topic mainly focuses on the sample processing,detection principle and process in the process of a variety of exosomes detection,as well as the automatic sampling of microfluidic chip.The main work is as follows:Firstly,based on the morphological characteristics of exosomes,the differences of surface markers and the characteristics of immunomagnetic microspheres,a set of microfluidic chips based on the combination of size control and magnetic field control was designed and manufactured to verify the functions of magnetic particles of different sizes.Based on microfluidics,simultaneous detection of Jurkat exosomes from oral ulcers and Cal-27 exosomes from oral cancer were performed.The detection limits for exosomes from oral ulcers and oral cancer were 8.5×10~4particles/m L and 4.45×10~5particles/m L,respectively,with high sensitivity.Combined with the self-designed automatic sampling device,the automatic sampling of the testing process is realized,and the control of the upper computer is realized.In the process of automatic injection device construction,combined with the actual demand,developed a crystal oscillator components quality testing system based on LabVIEW.Through the control of a variety of physical structures in the microfluidic chip,this project realized the simultaneous detection of oral cancer and oral ulcer exosomes,and built an automated sampling system to further improve the automation of exosome detection,which has certain clinical application value.
