Background and ObjectiveThe period of orthodontic treatment is as long as 2 to 3 years or even longer,and the long-term treatment will increase the risk of adverse reactions.Therefore,how to shorten the orthodontic treatment time and accelerate the orthodontic tooth movement are the concerns of the dental surgeon.The essential Tprocess of orthodontic tooth movement is the reconstruction of periodontal tissues,of which the alveolar bone is the key.Accelerating alveolar bone reconstruction can accelerate orthodontic tooth movement.Low-intensity pulsed ultrasound can exert mechanical force stimulation on alveolar bone marrow mesenchymal stem cells and affect their early differentiation ability.However,the mechanism underlying remains to be further studied.Therefore,this experiment intends to investigate the effect of low-intensity pulsed ultrasound stimulation on the osteogenic differentiation of rats alveolar bone marrow mesenchymal stem cells,and to explore the role played by the FAK-PI3K/ AKT signaling pathway.Materials and MethodsIsolation and culture of rat alveolar bone marrow mesenchymal stem cells.Stimulate cells with low-intensity pulsed ultrasound stimulation of different intensity parameters in vitro.Explore cell proliferation activity、osteogentic gene m RNA expression and alkaline phosphatase staining to evaluate the effect of ultrasound on the early stage of cell osteogentic stage.Find out the optimal intensity of ultrasound to promote the proliferation and differentiation of rat alveolar bone marrow mesenchymal stem cells.Western blot was used to detect FAK-PI3 K / AKT signaling pathway protein expression after ultrasound stimulation.The alveolar bone marrow mesenchymal stem cells were pretreated with signal pathway-specific chemical inhibitors.After ultrasound stimulation,the osteogentic gene m RNA expression was examined to verify the regulatory role of the FAK-PI3 K / AKT signaling pathway.Results In vitro low-intensity pulsed ultrasound in vitro can promote osteogenic differentiation of rat mandible bone marrow mesenchymal stem cells,and the FAK-PI3 K / AKT pathway plays an important role in it.It converts the mechanical signals of ultrasound into intracellular biological signals,thereby regulating the osteogenic differentiation process of cells. |
