| Objectie:To explore the effect of Rhizoma et Radix Notopterygii(qiang huo)water extract on the liver tissue MDA content,SOD activity and GSH-Px activity of ischemia-reperfusion injury model rats and the mechanism of reducing liver injury.Methods:54 6-week-old SPF male SD rats were divided into different groups randomly:blank group(B group,B),simple model group(Model group,M),and Qianghuo water extract pretreatment group(Qiang huo group,QH).There are eighteen animals in each group.The blank group was divided into three groups according to ischemia forty-five minutes,ischemia forty-five minutes reperfusion thirty minutes,and ischemia forty-five minutes reperfusion sixty minutes,with six animals in each group,namely I45 group(I45),I45R30 group(I45R30)and I45R60 group(I45R60).M group and QH group were divided into groups of I45,I45R30 and I45R60 according to the ischemia reperfusion duration of the B group.The rats were all reared adaptively for one week.After adaptively rearing,the rats in the QH group were gavaged with the Qianghuo water extract on time daily for one week.The rat model is only aimed at the M and QH groups.The modified modeling method:Routinely anesthetize the rat.After the anesthesia is successful,open the laparotomy and bluntly separated the left middle lobe of the liver,and the root of the left middle lobe of the liver was clamped with non-invasive arterial clamps.According to the needs of the experiment,remove the non-destructive arterial clip and allow the blood to reperfuse the liver for thirty minutes and sixty minutes.Afer thirty minutes and sixty minutes of reperfusion,blood was collected from the inferior vena cava of the rat,and the lobe of the modeled liver was obstained.Observation of the pathological changes of modeled liver lobes under light microscope after hematoxylin-yin red staining.Test rats serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),liver tissue malondialdehyde(MDA)content,superoxide dismutase(SOD)activity,glutathione Peroxidase(GSH-Px)activity level.Results(1)Transaminase levels in each group:compared with group B,the blood ALT and AST levels of rats in group M increased(P<0.01),indicating that the model was successful.The liver enzyme level reached the highest level of the three time points at60 minutes after reperfusion.Compared with the model group of rats,the serum ALT level of rats in the QH group treated with the water extract of Qianghuo was decreased 45 minutes after operation(I45 immediately),75 minutes after operation(I45R30 immediately)and 105 minutes after operation(I45R60immediately)(P<0.01);the serum AST level of rats in the QH group was decreased 75minutes(I45R30 immediately)and 105 minutes(I45R60 immediately)after surgery(P<0.01).(2)MDA level of liver tissue in each group:Compared with rats in the blank group,the MDA content in liver tissue of rats in the model group increased at 45 minutes,75 minutes and 105 minutes after surgery(P<0.01),with the highest level at 60 minutes after reperfusion.Compared with rats in the model group,the MDA content in liver tissue of rats in the Qianghuo water extract pretreatment group decreased at 45 minutes,75 minutes and 105 minutes after surgery(P<0.01),but compared with rats in the blank group,the liver of rats in the QH group tissue MDA content has increased(P<0.01).(3)SOD and GSH-Px levels in liver tissues of each group:Compared with the blank group,the SOD activity and GSH-Px activity levels in the liver tissue of the model group decreased at 45 minutes,75 minutes,105 minutes after surgery(P<0.01),and reached the lowest point at three points at 60 minutes after reperfusion.Compared with the model group,the SOD activity level in the liver tissue of rats in the Qianghuo water extract pretreatment group increased at 45 minutes and105 minutes after surgery(P<0.01),and the GSH-Px activity level increased at 75 minutes and 105 minutes after surgery(P<0.01).(4)Observation after HE staining of modeled liver lobe section:the hepatocyte cords in group B are neatly arranged,the liver lobules are clearly structured,and there is no blood stasis and no inflammation cell infiltration in the central venous area.Following the process of ischemia and reperfusion,rats in group M showed changes from cytoplasmic porosity,cell edema,ballooning,to cell necrosis,to obvious dilation and blood stasis of the liver sinusoids,and pathological changes in the area of flaky cell necrosis.Among the groups,the injury is the most serious at 60 minutes after reperfusion.The pathological damage of the QH group was improved compared with the M group at the corresponding time points.ConclusionThe pretreatment of Qianghuo water extract can reduce the level of transaminase in rats with hepatic ischemia-reperfusion injury,increase the activity level of SOD and GSH-Px in liver tissue,and reduce the production of MDA in liver tissue,reduce inflammation and oxidative stess damage.The results of HE staining pathological section of modeled liver lobes showed that the pretreatment of the Qianghuo water extract can reduce the degree of liver tissue pathological damage in HIRI rats.It can be inferred that the Qianghuo water extract intervention can antagonize HIRI and exert a liver protective effect. |
