Objective: In this study,the expression and significance of toll-like receptor-2(TLR-2),cysteinyl aspartate specific proteinase-3(Caspase-3)and matrix metalloproteinase-3(MMP-3)in the fetal membranes of parturient with premature rupture of membranes(PROM)in Xining area were preliminarily discussed,so as to provide certain experimental basis for the prevention and treatment of PROM in Xining area.Methods:(1)30 cases of the fetal membrane tissues samples of women with PROM and 30 cases of the fetal membrane tissues samples of women with normal pregnancy during the same period were randomly selected from the Affiliated Hospital of Qinghai University from September 2019 to August 2020.Both groups gave informed consent.There were no other obstetric complications in the two groups.(2)The localization and expression of TLR-2,Caspase-3 and MMP-3 in the fetal membrane tissues of the PROM group and the normal pregnant group were detected by immunohistochemical method.The expression of TLR-2,Caspase-3 and MMP-3in the fetal membrane tissue of the two groups were detected by Western blot.(3)The independent two-sample t-test was used to analyze whether the expression of TLR-2,Caspase-3 and MMP-3 were different in the fetal membrane tissue between the two groups in this area.The correlation between the two variables was analyzed by Pearson correlation.Results:(1)TLR-2,Caspase-3 and MMP-3 were mainly located in the cytoplasm of amniotic epithelial cells and chorionic trophoblast cells,and the localization of two groups is basically the same.The average optical density value of TLR-2,Caspase-3and MMP-3 in PROM group were higher than that in normal pregnancy group,and the difference was statistically significant(P<0.05).The relative expression levels of TLR-2,Caspase-3 and MMP-3 were higher than that in normal pregnancy group,and the difference was statistically significant(P<0.05).(2)Immunohistochemistry and Western blot detection showed that the expression level of TLR-2 was significantly positively correlated with that of MMP-3 in the fatal membrane tissues of PROM group(r=0.472,P<0.01;r=0.640,P<0.01),but no significant correlation was found between the two in the normal pregnancy group.(3)Immunohistochemistry and Western blot detection showed no significant correlation of the expression level of Caspase-3,the expression level of TLR-2 and MMP-3 in fetal membrane tissues of the two groups.Conclusions:(1)The localization of TLR-2,Caspase-3 and MMP-3 in the fetal membrane tissue of the PROM group and the normal pregnant group in Xining area were basically the same.However,the expression of TLR-2,Caspase-3 and MMP-3in the fetal membrane tissue of the PROM group was increased.TLR-2,Caspase-3and MMP-3 may be related to the occurrence of PROM in this region.(2)The expression level of TLR-2 was significantly positively correlated with the expression level of MMP-3 in the fatal membrane tissues of the PROM group in Xining area,but no significant correlation was found between the two in the normal pregnancy group,suggesting that both of them may participate in the pathophysiological process of PROM in Xining area.(3)There was no significant correlation of the expression level of Caspase-3,the expression level of TLR-2 and MMP-3 in fetal membrane tissues of the PROM group and the normal pregnant group in Xining area.Consider it unrelatedly possible or the reasons for smaller samples.
