Background:Breast cancer is the most common malignant tumor in women,and its incidence is increasing year by year.Although chemotherapy and other comprehensive treatment methods have greatly improved the patient’s prognosis,its serious side effects have a huge impact on the patient’s quality of life.Therefore,it is urgent to find more effective and tolerable anti-cancer therapies.In recent years,traditional Chinese medicine has played an important role in the process of cancer treatment.Dandelion,as one of the anti-tumor members of traditional Chinese medicine,has obvious anti-cancer activity against malignant tumors such as the digestive tract.However,its influence on the malignant biology of breast cancer cells and related mechanisms are still unclear.Purpose:This study intends to observe the effect of dandelion flavonoids on the migration and invasion of breast cancer MCF-7 cells through in vitro experiments,and to preliminarily explore the relevant mechanisms of its regulation of MCF-7 cell metastasis,so as to provide new ideas for the treatment of breast cancer with Chinese medicine.Methods:Human breast cancer MCF-7 cells were cultured in vitro,and dandelion flavonoids(250 μmol/L,500 μmol/L)with different drug concentrations were prepared to act on MCF-7 cells.After 24 hours,the scratch test was used to observe the changes in cell migration ability;Transwell experiment was used to detect the effect of dandelion flavone on the invasion of breast cancer MCF-7 cells;real-time polymerase chain reaction(RT-PCR)was used to detect MMP-2,MMP-9,TIMP-in MCF-7 cells after treatment with dandelion flavone 1 and changes in TIMP-2 mRNA levels.Results:(1)Dandelion flavonoids have an inhibitory effect on the migration ability of MCF-7 cells.As the concentration increases,the inhibitory effect is enhanced(all P<0.05).(2)Transwell experiment showed that dandelion flavonoids inhibited the invasion ability of MCF-7 cells,and its inhibitory effect also increased with the increase of concentration(all P<0.05).(3)RT-PCR method detected after adding dandelion flavone treatment,the expression of MMP-2 and MMP-9 mRNA in MCF-7cells decreased with the increase of flavone concentration,and the expression of TIMP-1 and TIMP-2 mRNA increased with the increase of concentration.Increase(all P<0.05).Conclusions:Dandelion flavone may inhibit the migration and invasion of MCF-7cells on the expression of MMP-2,MMP-9,TIMP-1 and TIMP-2 level.
