Objective: To study the effect of acupoint catgut embedding combined with traditional chinese medicine raspberry on serum sex hormone level and inflammatory factors in PCOS-IR model rats,to explore its molecular mechanism through TXNIP-NLRP3 signaling pathway,and to provide a new scheme and experimental basis for clinical treatment.Methods: 70 SD rats(females),select 10 normal groups on the basis of the random number and table method,60 models,PCOS-IR animal model was induced by subcutaneous injection of DHEA,continuous appearance of keratinized epithelial cells and HOMA-IR>2.8 As a mark of success.The results were 53,randomly selected 50 successful models,according to the random number table method divided into model group(n=10),acupoint catgut embedding group,for short,catgut embedding group(n=10),metformin group,for short,positive control group(n=10),traditional chinese medicine raspberry group,for short,raspberry group(n=10)and acupoint catgut embedding combined with traditional chinese medicine raspberry group,for short,the joint group(n=10).According to each group of treatment plan a total of 28 days.HE staining to observe the changes of ovarian histomorphology,ELISA detection of serum FINS、CRP、LH、SHBG and T,The HOMA-IR and FAI values are calculated by relevant formulas;W B and RT-PCR techniques were used to gauging the protein and m RNA expression levels of ovarian tissue TXNIP、NLRP3、ASC、P-caspase1、C-caspase1 and IRS-1.Results:1.DHEA subcutaneous injection,the vaginal epithelial cells in the model group were continuously keratinized;the FPG、FINS and HOMA-IR were markedly increased than those in the normal group(P<0.01);the number of corpus luteum in ovarian tissue decreased,the number of follicles at all levels decreased,the number of atresia follicles increased.2.In contrast with the model group,the ovarian morphology of each intervention group was improved to varying degrees,the corpus luteum increased,the vesicles and atresia follicles decreased,the granulosa cells were arranged regularly,especially the number and development of follicles at all levels in the combined group were close to normal,no cystic change was found in follicles.3.Compared with the model group,the LH content of raspberry group was down-regulated(P>0.05),and the serum FPG、FINS、CRP、LH、T content and HOMA-IR、FAI value of other treatment groups were down-regulated(P<0.01)and SHBG up-regulated(P<0.05).4.Compared with the normal group,in model group,the content of TXNIP、NLRP3、ASC、P-caspase1、C-caspase1 protein and m RNA were up-regulated(P<0.01),and the IRS-1 protein and m RNA were downregulated marked(P<0.01);compared to the model group,in positive control group and combined group,the content of TXNIP、NLRP3、ASC、P-caspase1、C-caspase1 protein and m RNA were decrecing(P<0.01),IRS-1 protein and m RNA expression levels were upregulated(P<0.01);in catgut embedding group,the expression levels of NLRP3、ASC、C-caspase1 protein was decrecing(P< 0.05),the expression level of TXNIP、P-caspase1 protein was trend down(P>0.05),C-caspase1 m RNA expression levels down-regulated(P<0.05),the expression level m RNA other factors was trend down(P>0.05),IRS-1 protein expression levels up-regulation(P<0.05),IRS-1m RNA expression levels was an upward trend(P>0.05);the expression of TXNIP、NLRP3、ASC 、 P-caspase1 protein in the raspberry group was upward trend(P>0.05),C-caspase1 protein and m RNA expression decrecing(P<0.05),the expression level m RNA other factors was trend down(P>0.05),IRS-1 protein expression levels up-regulated(P<0.05),IRS-1m RNA expression levels upward trend(P>0.05).Conclusion:1.DHEA modeling method can successfully induce PCOS-IR rat model with chronic inflammation.2.The intervention of each group,the level of hormone,inflammatory factor and insulin resistance in rats was down-regulated,and the morphology of ovarian tissue was improved to varying degrees,indicating that the combination of acupoint catgut embedding and traditional chinese medicine raspberry and its use alone had therapeutic effect on PCOS-IR and its chronic inflammation,but the combined application group was better.3.The expression of TXNIP、NLRP3、ASC、P-caspase-1、C-caspase-1 factors in ovarian tissue of model group was up-regulated,indicating that PCOS-IR microinflammation changes were related to the TINIP-NLRP3 signaling pathway.The above factors were down-regulated and IRS-1 expression up-regulated in ovary after treatment with acupoint catgut embedding combined with traditional chinese medicine raspberry.It may improve its inflammatory state by down-regulating androgen level,improving insulin resistance,and then inhibiting the activation of TXNIP-NLRP3 signaling pathway.
