Objective:In the hippocampus of rats miR-299b-3p_R+5 as a research point,find out the associated with diabetes and depression are common expressed genes,speculated that may involve regulatory mechanism between each other,on the basis of discusses radix bupleuri add keel oyster soup in treating diabetic depression may be the mechanism of action,for diabetic depression research provides a new Angle of thinking.Method:50 male SD rats were randomly divided into two groups: 10 rats and 40 rats.10 rats were divided into blank group and fed with normal diet.40 were made into a module.The model group was fed with high-sugar and high-fat diets,and after 4weeks,streptozotocin(STZ)was injected intraperitoneally to prepare the diabetes model.The model rats were selected to prepare the model of combined depression by chronic unpredictable stimulation(CUMS)and cage-fed alone.The open field test,swimming test and tail suspension test were used to evaluate the behavior of rats.Finally,30 rats qualified for behavioral test were selected and randomly divided into 3 groups: Bupleurum and Longgu oyster decoction group,fluoxetine group and model group.The body weight and blood glucose of rats were measured and recorded before,4 weeks and 8 weeks after administration.Eight weeks after administration,the samples were collected,and total RNA was isolated from the hippocampus of rats.Real-time PCR was used to detect the expression level of miRNA in the hippocampus,and a c RNA library of RNA molecules was established.Interested miRNAs were selected to analyze the differentially expressed genes.Result:1.Weight measurement: The weight of rats in each group increased steadily before drug treatment,and the weight growth showed an unstable trend after modeling.After drug administration,the weight of Bupleurum and Longgu Oyster Decoction group increased compared with the model group,with statistical significance(P < 0.05).There was no significant difference in body weight between fluoxetine group and model group(P > 0.05).2.Behavioral test: the number of activities in fluoxetine group and Bupleurum and Longgu Oyster Decoction group was increased compared with the blank group,with statistical difference(P < 0.05).3.miRNA expression: there was no significant difference between the Bupleurum and Longgu Oyster Decoction group and the fluoxetine group(P > 0.05).4.The enrichment analysis of differential miRNA target genes in the original data indicated that Prkca gene regulates the related pathways of diabetes and depression downstream,and may regulate the co-morbidity of diabetes and depression through circadian rhythm.Conclusion:1.The modeling mode of diabetes combined with depression made the weight of rats increase first and then decrease,and the weight fluctuated greatly,which was similar to the state of diabetes in humans,and may be related to the hormone disorder and blood glucose instability of diabetes itself.2.The treatment of Bupleurum and Longgu Oyster Decoction can increase the total distance of voluntary activity of rats,which can be considered as a positive indicator for the improvement of depressive behavior.3.Bupleurum and Longgu Oyster Decoction had no effect on blood glucose level of rats,suggesting that Bupleurum and Longgu Oyster Decoction may have no effect on blood glucose at both positive and negative feedback levels.4.Prkca gene may lead to the imbalance of the upstream and downstream pathways through the circadian rhythm disturbance,leading to the imbalance of hormones and blood sugar,and indirectly affect the function of the nervous system,which may be the co-pathogenic factor of diabetes combined with depression. |
