Objective:To study the effects of Bupleurum plus Longgu Oyster Decoction on behavior,body weight and expression of neurotransmitter DA in hippocampus of depressed rats,and to explore the anti-depression mechanism of Bupleurum plus Longgu Oyster Decoction.Method:A total of 70 SPF male SD rats were randomly divided into two groups:one group had 10 rats and the other group had 60 rats.A group of10 birds was numbered asⅠ,which was the blank group and given normal feeding.A total of 60 rats in group II were fed normally and Depression models were developed under complex conditions under the influence of multiple factors.The depression degree of rats was measured by tail-hanging,open-field,forced swimming test and animal behavior software.After screening theⅡgroup,the rats meeting the standard were randomly divided into three groups(model group,fluoxetine group and Bupleurum plus Longgu oyster decoction group).The body weight of each group was recorded every week,and the behavior test was conducted at 0,4 and 8 weeks after administration of medicine.The expression of DA in hippocampus was detected by immunohistochemistry.Real-time PCR was used to detect the mRNA expression of D2 in hippocampus.He staining was used to observe the changes of cells in the hippocampus.Result:1.Effect on weight:After the completion of modeling,the body weight of blank group was significantly higher than that of model group(P<0.05).After 4 and6 weeks of intragastric administration,the body weight of each group increased,but there was no significant difference between groups(P>0.05).After 8 weeks of intragastric administration,There was no significant change in the trend of weight gain,there was no significant difference between the administration group and the model group(P>0.05).2.Behavioral testing:Changes in swimming and floating time of rats:before drug intervention,the immobile floating time of model group was significantly longer than that of blank group(P<0.05).After 4 and 8 weeks of treatment,the immovable floating time of fluoxetine group and Bupleurum plus Longgu oyster group was significantly shortened compared with model group,and the difference was significant(P<0.05).Changes in the immobility time of tail suspension in rats:before drug intervention,the immobility time of tail suspension in model group was significantly longer than that in blank group,with statistical significance(P<0.05).After 4 weeks of intragastrium,the immobility time of tail suspension in fluoxetine group was significantly shorter than that in model group(P<0.05).After 8 weeks of intervention treatment,the immobility time of tail suspension in fluoxetine group and Chaihu+Longgu oyster group was significantly shorter than that in model group(P<0.05).Changes of open field experiment in rats:compared with the blank group before drug intervention,the total walking distance,wall climbing and standing time,and rest time were significantly decreased in the model group,with statistical significance(P<0.05).After 4 weeks of treatment,the total distance walking in fluoxetine group was significantly increased compared with the model group(P<0.05).Compared with model group,the rest time was significantly decreased in Bupleurum plus Longgu oyster decoction group and fluoxetine group(P<0.05).There were no significant differences in wall climbing time and standing time between the Bupleurum plus Longgu oyster Decoction group and fluoxetine group(P>0.05).After 8 weeks of drug intervention,the total walking distance of rats in the Bupleurum plus Longgu oyster Decoction group and fluoxetine group was significantly increased compared with the model group(P<0.05).Compared with model group,the rest time of fluoxetine group and Bupleurum plus Longgu oyster group was significantly decreased(P<0.05).Compared with model group,there were no significant differences in wall climbing and standing time between Bupleurum plus Longgu oyster group and fluoxetine group(P>0.05).3.Real-time PCR detection D2mRNA mRNA expression:After 8 weeks of drug intervention,the expression of D2mRNA in fluoxetine group and Bupleurum plus Longgu oyster decoction group was significantly increased compared with model group(P<0.05).4.The expression of immunohistochemical method to detect DA:After 8 weeks of drug intervention,compared with model group,the expression of DA was significantly increased in Bupleurum plus Longgu oyster Decoction group and fluoxetine group,with statistical significance(P<0.05).5.HE staining of the hippocampus:The morphology of the vertebral cells in the hippocampus of the model group was irregular and scattered,with nuclear pyknosis.After 8weeks of drug intervention,the vertebral cells in the hippocampus of rats in the Bupleurum+Longgu oyster Decoction group and fluoxetine group were arranged regularly and distributed evenly,with occasional deep taining of nuclei and larger cell bodies.Conclusion:1.The use of CUMS combined with isolated cultivation model may slow down the growth rate of body weight of rats,while the treatment of Bupleurum and Longgu Oyster Decoction has no effect on the change rend of body weight.2.Bupleurum and Dragon Bone Oyster Decoction can increase the umber and time of autonomic activity,reduce the degree of stupidity and improve the depressive behavior of the depressed rats.3.Bupleurum and Longgu Oyster Decoction can increase the D2RNA concentration in the hippocampus of depressed rats,and increase the expression of DA protein,which may be the anti-depression echanism.4.Bupleurum and dragon bone oyster soup can improve the isordered arrangement and mutated structure of the vertebral cells in the hippocampus of depressed rats,and then repair the plasticity of neurons. |
