| Objective: In this study,the levels of serum neurofilament light chain(NFL)and interleukin 6(IL-6)in patients with Parkinson’s disease(PD)were detected to explore the severity of serum NFL,IL-6,and PD disease.The correlation between the degree and cognitive dysfunction provides a basis for the further study of PD peripheral serum biomarkers.Methods:Ninety-nine researchers were enrolled as research subjects,63 patients with primary PD were enrolled as the case group,and 33 healthy subjects were enrolled as the control group.In addition,according to HY staging(Hoehn-Yahr grading,HY grading),the cases were divided into two subgroups: early Parkinson’s disease and middle and late Parkinson’s disease.According to the Mini-mental State Examination(MMSE)score,The case groups are divided into two subgroups: None cognitive impairment in PD(PD-NC)group and Parkinson Disease-cognitive impairment(PD-CI).Collect blood samples from all the case groups and control groups,and collect general information such as age,gender,personal history,and family history.In the case group,all investigators were evaluated by H-Y staging and MMSE score.Enzyme-linked immunosorbent assay(ELSIA)was used to detect serum IL-6 and NFL levels in the case group and the control group,and the differences in the above indicators between the case group and the control group and the subgroups of the case group were analyzed and clinical Feature correlation.Results:1.Through the analysis of the serum NFL levels of all researchers,the serum NFL levels of the subjects in the PD group were significantly higher than those in the control group,and the difference was statistically significant(P<0.05).In the two subgroups of the early Parkinson’s disease group and the middle-advanced Parkinson’s disease group,the serum NFL level of the subjects in the middle-advanced group was higher than that of the early-stage group,and the difference was statistically significant(P<0.05).In the two subgroups of the PD-NC group and the PD-CI group,the serum NFL level of the subjects in the PD-CI group was higher than that of the PD-NC group,and the difference was statistically significant(P<0.05).The correlation analysis between serum NFL level and PD patients’ H-Y staging,MMSE score and age found that the relationship between PD patients’ serum NFL level and H-Y staging was positively correlated(P<0.05).The higher the H-Y staging,the higher the serum NFL level.The serum NFL level of PD patients was negatively correlated with the MMSE score results(P<0.05).The lower the MMSE score,the higher the serum NFL level.There was no correlation between the serum NFL level of PD patients and gender(P>0.05).In the analysis of the value of serum NFL level predicting the occurrence of PD,the area under the curve(ROC area under the curve,AUC)of the receiver operating characteristic(ROC)curve(ROC)of serum NFL predicting the occurrence of PD is 0.944,and the P value is less than 0.001,statistically significant(P<0.05).2.Through the analysis of the serum IL-6 levels of all researchers,the serum IL-6 levels of the subjects in the PD group were significantly higher than those in the control group,and the difference was statistically significant(P<0.05).There was no significant difference in serum IL-6 levels in the two subgroups of the early Parkinson’s disease group and the middle-advanced Parkinson’s disease group(P>0.05;in the two subgroups of the PD-NC group and the PD-CI group,serum IL-6 levels There was no significant difference in IL-6 levels(P>0.05).The correlation analysis between serum IL-6 level and H-Y staging,MMSE score and age of PD patients found that there was no correlation between serum IL-6 level and H-Y staging MMSE score,and age of PD patients(all P>0.05).In the analysis of the value of serum IL-6 levels in predicting the occurrence of PD,the area under the ROC curve(AUC)of serum IL-6 predicting the occurrence of PD was 0.909,with a P value <0.001,which was statistically significant(P<0.05).Conclusions: 1.The serum NFL level of PD-CI patients increases,and the worse the cognitive function is,the higher the serum NFL level;the serum IL-6 level of PD patients has no significant correlation with PD-CI.2.Serum NFL and IL-6 levels in PD patients are elevated,and both may be involved in the pathogenesis of PD.3.With the change of H-Y staging,the serum NFL level of patients with advanced Parkinson’s disease is higher than that of patients with early Parkinson’s disease,suggesting that the serum NFL level reflects the severity of PD to a certain extent. |
