| Objective: Observe the expression of α-synuclein gene in gastric cancer tissues,analyze its relationship with the clinical staging and pathological differentiation of gastric cancer,and prompt further exploration of effective methods for the prevention and treatment of gastric cancer.Methods: The subjects of the study were patients with advanced gastric cancer who were admitted to the Department of Gastrointestinal Surgery of Guilin Medical College Affiliated Hospital from August 2018 to December 2018.Fresh gastric cancer tissue and adjacent tissues were taken during the operation and cryopreserved in liquid nitrogen(preoperative Gastric cancer was diagnosed by pathological results of gastroscopy or routine pathological diagnosis after surgery),and then divided into different subgroups according to clinical stage and tissue differentiation.Perform immunohistochemical staining on cancerous tissues and adjacent tissues,and integrate them;Extract RNA from the tissues,perform reverse transcription,and then use Real-time Quantitative PCR Detecting System to determine the expression of gene m RNA.Analyze its expression differences with different clinical stages and different degrees of differentiation.Results: Collected 50 patients with gastric cancer,including 37 male patients and 13 female patients;38 patients 60 years old or more,12 patients younger than 60 years old;the number of patients with clinical stages ofⅠ,Ⅱ,andⅢ were 6 cases,6 cases and 38 cases;11 cases were pathologically differentiated into well-differentiated cases,39 cases were poorly differentiated.1.The results of immunohistochemical score analysis showed that among the50 patients,the number of high expression cases in cancer tissue were 14 cases,and the number of low expression cases were 36 cases,the number of high expression cases in adjacent tissues were 29 cases,and the number of low expression cases were 21 cases.It is statistically significant(χ2=9.180,P=0.0024);In different clinical stages,the number of high expression cases in stage Ⅰ,Ⅱ,and Ⅲ gastric cancer tissues were 5,2 and 7 respectively,and the number of low expression cases were 1 case respectively,4 cases and 31 cases,the difference was statistically significant(χ2=10.93,P=0.0042);The immunohistochemical results of the well-differentiated group showed that there were 8 cases in the well-differentiated group,3 cases in the low-expression group,and 6 cases in the poor-differentiation group,33 cases in the low-expression grope,the difference was statistically significant(χ2=13.99,P=0.0002).2.Real-Time PCR counting and statistical analysis results showed that the number of high expression cases in cancer tissue were 15 cases,and the number of low expression cases were 26 cases;the number of high expression cases in adjacent tissues were 25 cases,and the number of low expression cases were 16 cases.The difference was statistically significant(χ2=5.321,P=0.0211);In different clinical stages,the number of high expression cases in stage Ⅰ,Ⅱ,and Ⅲgastric cancer tissues were 4,2 and 9 cases respectively,and the number of low expression cases were 1,3 and 22 cases,respectively,the difference was statistically significant(χ2=4.498,P=0.0339);In the highly differentiated group,7 had high expression in the well differentiated group,2 had low expression,8had high expression in the poorly differentiated group,and 24 had low expression.The difference was statistically significant(χ2=8.434,P=0.0037).The results of quantitative statistical analysis showed that the mean±standard deviation of tumor and adjacent tissues were 0.78±0.54 and 4.02±3.57,respectively,and the difference was statistically significant(t=3.127,P=0.0030);In different clinical stages,the mean ± standard deviations of stageⅠ,Ⅱ,and Ⅲ were 5.161±4.14,0.680±0.53,0.364±0.32,respectively,and the difference is statistically significant(F=5.236,P=0.017);Among different levels of differentiation,the mean ± standard were 16.55±10.12 and 0.703±0.42,respectively,and the differences were statistically significant(t=3.367,P=0.0025).3.In general data statistics,the data were compared with the differences between the groups through the χ2 test,and the results showed that the differences between the groups such as gender,age,preoperative hemoglobin level,preoperative albumin level,etc.were not statistically significant(P>0.05).Conclusion: 1.The expression of SNCA in gastric cancer tissues is significantly lower than that of adjacent tissues,suggesting that the down-regulation of SNCA gene may be involved in the occurrence and development of gastric cancer;2.SNCA is significantly low expressed in poorly differentiated gastric cancer tissues,suggesting that the detection of SNCA gene expression may help determine the severity of gastric cancer patients;3.The expression of SNCA in stage Ⅲ gastric cancer tissue is significantly lower than that in stageⅠ,suggesting that the inactivation of SNCA gene may lead to aggravation of gastric cancer patients. |
