Objective:The purpose of this study was to explore whether the sensitivity of gastric cancer cells with diverse differentiation to hyperthermia is the same or not by studying the proliferation,invasiveness and expression of Hsp70 protein in three kinds of gastric cancer cells,and to provide a theoretical basis for individualized hyperthermia of gastric cancer patients.And we will initially explore the expression of Hsp70 protein in the three kinds of gastric cancer cells with different differentiation.Methods:There are three diverse differentiated gastric cancer cell lines.They are well differentiated AGS cells,moderately differentiated SGC7901 cells,and poorly differentiated BGC-823 cells.The cells were cultured to logarithmic growth phase when the cell lines were taken for experimental study.All the three different cells were divided into hyperthermia group and normal temperature group respectively.After heat treatment at 43?5%CO2 for 1 hour,the cells in the hyperthermia group were incubated at 37?5%CO2 for 23 hours,while the cells in the normal temperature group were cultured continuously at 37?5%CO2for 24 hours.The corresponding treated cells were tested as follows:1.MTT test was operated to analyze the proliferation ability of each cell line;2.Transwell chamber invasion test was used to study invasiveness of cells;3.Western-blot test was used to detect the expression of Hsp70protein.Results:1.MTT test showed that,compared with the normal temperature group,OD values of each group in hyperthermia were all decreased?P<0.05?.The relationship between the inhibitory rate of 24th and 48th of hyperthermia on the proliferation of different cancer cell lines was as follows:AGS>SGC7901>BGC-823?P<0.05?;and at the time of 72th,the inhibition rates of hyperthermia on cells were as follows:SGC7901>AGS?P<0.05?,SGC7901>BGC-823?P<0.05?,BGC-823>AGS?P>0.05?.2.The results of Transwell chamber invasion test showed that,compared with the normal temperature group,the invasive cell number of hyperthermia group decreased?P<0.05?.The relationship between the inhibitory rate of hyperthermia on the invasion of different gastric cancer cell lines were as follows:AGS>SGC7901>BGC-823?P<0.05?.3.The results of Western-blot were as follows.Compared with the normal temperature group,the expression of Hsp70 protein in hyperthermia group was significantly up-regulated?P<0.05?.And the expression of Hsp70 in each cell line after hyperthermia is as follows:SGC7901>BGC-823>AGS?P<0.05?.Conclusions:1.Hyperthermia can inhibit the ability of proliferation and invasiveness of all the gastric cancer cell lines.And the higher degree of differentiation the cell line is,the stronger repressive effect of hyperthermia is.2.Hyperthermia could significantly up-regulate the expression of Hsp70 protein in the three different gastric cancer cell lines.
