| Objective: To investigate the effect of aconite injection on the content of Aggrecan in the joint fluid of rabbit model of KOA,and to explore the therapeutic effect of aconite injection on KOA.Methods: twenty-six big-eared white rabbits were randomly divided into two groups:blank group and model group.Group A was blank group(without any treatment)with A total of 7 rabbits.Group B was the model group(given different manual treatment),a total of19 rats.In order to ensure the reliability and scientificity of modeling,The model group was made by papain induction modeling method.After the injection of the modeling reagent The white rabbits were randomly divided into three groups : model group(B),Zhengqing Fengtongning group(C)and aconitum injection group(D),and Each group of six.Group B was given 0.2 m L of normal saline,Group C was given 0.2 m L of Zhengqing Fengtongning injection,and Group D was given 0.2 m L of aconitum injection.After the completion of the administration.Finally,all the experimental rabbits were killed,and the rabbits were dissected to observe the general condition of the joints and the changes of the cartilage,and detailed records and analysis were made.The content of Aggrecan in the sample joint fluid was determined by ELISA;The pathological sections stained with HE were observed under microscope;the naked eye observes the changes in knee joints and their cartilage.Result:1.naked eye observation: The articular cartilage in the blank group basically had no change,but the model group had the largest change.The articular cartilage was rough with osteophyte formation,and the color was mostly dark yellow.The defect and damage of Zhengqing Fengtongning injection group were smaller than that of model group,and the articular cartilage of aconitum injection group was similar to that of blank group.The content of Aggrecan in the knee joint fluid of rabbits in aconitum injection group and Zhengqing Fengtongning group was lower than that in blank group,with statistical significance(P < 0.05).Aggrecan content in rabbit knee joint fluid of aconitum injection group was higher than that of Zhengqing Fengtongning group,with statistical significance(P< 0.05).2.Microscopy observation of chondrocytes:the morphology and structure of the slice cells in the blank group were normal,while the apoptosis and loss of chondrocytes in the model group were obvious.The Zhengqing Fengtongning injection group had apoptotic cells like white bubbles,and the morphological and structural disorder was not obvious.3.ELISA results: the content of Aggrecan in rabbit knee joint fluid in model group was significantly lower than that in blank group,with statistical significance(P(27)0.05);the content of Aggrecan in rabbit knee joint fluid in aconitum injection group,zheng qing feng tong ning group was higher than that in model group,with statistical significance(P(27)0.05).Aggrecan in rabbit joint fluid in aconitum injection group was higher than that in control group,with statistical significance(P(27)0.05).Conclusion:1.Aconitum injection can promote the content of Aggrecan in knee joint fluid,and has obvious therapeutic effect on knee osteoarthritis.2.Aconitum injection can inhibit the apoptosis of chondrocytes and protect chondrocytes.3.Aconitum injection can prevent the destruction of articular cartilage,protect the cartilage and delay the development of the disease. |
