| BACKGROUND:Triple negative breast cancer(TNBC)is a highly invasive breast cancer,which is mainly treated by surgery and chemotherapy.The combination chemotherapy is also a treatment trend which choose different mechanism of drugs,to reach better clinical effect meanwhile reduce the side effects and drug resistance.OBJECTIVE: To investigate if the Pseudomonas aeruginosa injection(PAMSHA)combined with paclitaxel can inhibit TNBC cells and its possible mechanism.METHODS:(1)The inhibition of PA-MSHA,paclitaxel and the combination two on TNBC cell proliferation were evaluated by CCK8 assay.(2)The effect of PAMSHA on TNBC cell migration was evaluated by scratch test.(3)The effects of PAMSHA on the cell cycle and apoptosis of TNBC were detected via flow cytometry(FCM).PA-MSHA,paclitaxel and the combination two on cell apoptosis were compared.(4)The influence of PA-MSHA,paclitaxel and the combination on the expression of BAX,BCL-2,c-Myc,NF-κB and CIP2 A were detected by Western Blot and q RT-PCR.RESULTS:(1)CCK8 assay indicated that PA-MSHA,paclitaxel and the combination therapy all could inhibit the proliferation of TNBC cells,but the combination therapy had better effect than single therapy(P < 0.05).(2)Compared with the control group,the scratch healing rate of TNBC cells treated with PA-MSHA for24 h in the experimental group was lower(P < 0.05),and the migration of TNBC cells was significantly inhibited.(3)FCM results showed that the apoptosis rate of experimental group was higher than single therapy.In addition,the cell cycle arrested at the G0/G1 phase by the treatment of PA-MSHA.(4)Western Blot and q RT-PCR results showed that PA-MSHA,paclitaxel and the combination therapy could increase the expression of BAX,and decrease the expression of BCL-2,c-Myc,NF-κB and CIP2A(P < 0.05).Moreover,the down-regulation of target gene was more significant in combination therapy group.CONCLUSIONS: PA-MSHA can effectively inhibit the proliferation and migration of TNBC cells,and induce apoptosis and cycle arrest of TNBC cells.The mechanism of apoptosis may be related to BAX and BCL-2,and the combination therapy has more significant effect.PA-MSHA combined with paclitaxel can enhance the tumor inhibition effect of chemotherapy drugs,and its molecular mechanism may be related to the down-regulation of the expression of oncogenes NF-κB,c-Myc and CIP2 A by PA-MSHA. |
