Objective:With the in-depth study of microRNAs,it has been discovered that abnormally elevated or decreased microRNAs are not only closely related to the proliferation,invasion,metastasis,and radiotherapy and chemotherapy resistance of malignant tumors,but also play an important biological regulatory function.This study aims to clarify the biological function of hsa-miR-7-5p in head and neck squamous cell carcinoma(HNSCC).Methods:Analyze the relevant data in the TCGA database to obtain the expression of hsa-miR-7-5p in HNSCC,and draw the survival curve of patients with differentially expressed hsamiR-7-5p in Kaplan-Meier Plotter.The expression of hsa-miR-7-5p was detected by qPCR;the functions of hsa-miR-7-5p were verified by cck8,clone formation,transwell(invasion and migration),flow cytometry detection cycle and apoptosis,when we overexpressed hsamiR-7-5p through transfecting mimic of hsa-miR-7-5p and downregulated hsa-miR-7-5p through transfecting inhibitor of hsa-miR-7-5p.Results:Hsa-miR-7-5p is up-regulated in head and neck squamous cell carcinoma tissues and cell lines,especially in laryngeal squamous cell carcinoma,this molecule is negatively correlated with patient prognosis.In vitro cell experiments show that overexpression of hsamiR-7-5p in CAL-27,Fa Du,Detroit 562,AMC-HN-8 and FD-LSC-1 cells can enhance the proliferation and plate cloning ability of head and neck squamous cell carcinoma.After overexpressing hsa-miR-7-5p in CAL-27,Fa Du,Detroit 562,and FD-LSC-1 cells,the invasion and migration ability was significantly enhanced;on the contrary,inhibiting the expression of hsa-miR-7-5p can inhibit tumor cell proliferation,plate cloning,invasion and migration capabilities.However,in vitro changes in the expression of hsa-miR-7-5p have no statistically significant effects on the cycle and apoptosis of head and neck squamous cell carcinoma.Conclusion:The expression of hsa-miR-7-5p is high in HNSCC,which can evidently promote proliferation,invasion and migration of HNSCC,and play a key role in the occurrence and progression of tumors.Thus,hsa-miR-7-5p may play an oncogenic role in HNSCC,and hsamiR-7-5p can be used as a potential prognostic marker and therapeutic target for HNSCC.
