Objective:This study by observing the ridge electric acupuncture combined with neural mobilization to conformity of sciatic nerve injury after shrinking gastrocnemius muscle cells apoptosis,muscle wet weight ratio,the influence of the Bcl-xL,Bad expression,clip ridge electric acupuncture combined with neural mobilization myocyte apoptosis inhibition of rabbit sciatic nerve injuries,delay the muscle wet weight than the decline speed of related regulatory mechanism,for clip ridge electric acupuncture combined with nerve manipulation treatment of nerve muscle atrophy.Methods:180 Japanese large eared white rabbits,half male and half female,were randomly divided into 5 groups:normal control group,model control group,nerve loosening group,Jiaji electroacupuncture group,Jiaji electroacupun-cture combined with nerve loosening group(combined group),36 rabbits in each group.Each group was divided into 3 subgroups with 12 animals in each subgroup according to time point(1,2,4 weeks after treatment).The rabbit model of acute sciatic nerve injury was established by sciatic nerve clamp.The normal control group did not receive any treatment,and the model control group did not receive any intervention treatment after modeling.The Jiaji electroacupuncture group,the nerve loosening group and the combina-tion group were respectively given the treatment of Jiaji electroacupuncture,nerve loosening and the combination of electroacupuncture and nerve loosening on the 3rd day after surgery.After 1,2 and 4 weeks of treatment,the wet weight of bilateral gastrocnemius muscle was measured and the wet weight ratio was calculated.TUNEL method was used to observe the apoptosis of gastrocnemius myocytes.The expression of Bcl-XL and Bad mRNA was observed by RT-PCR.The expression of Bcl-XL and Bad protein was observed by Western Blot.Results:1.Gastrocnemius wet-weight ratioThe wet/gain ratio of the normal control group was 1.At the three time points of week 1,2 and 4,the wet/gain ratio of all groups(except the normal control group)decreased continuously with time,and reached the lowest value at week 4.At the same time point,the wet to gain ratio of treatment group was lower than that of normal control group(P<0.05),higher than that of model control group(P<0.05),combined group was higher than that of nerve loosening group and jiaji electroacupuncture group(P<0.05);At the same time point,there was no significant difference between electroacupunct-ure group and nerve loosening group(P<0 05)..2.Tunel detection of apoptosis indexAt the time of 1,2 and 4 weeks,the apoptosis index of all groups(except the normal control group)increased continuously with time,and reached the highest value at 4 weeks.At the same time point,the apoptosis index of treatment group was higher than that of normal control group(P<0.05),and lower than that of model control at the same time point(P<0.05),and the combination group was lower than that of nerve loosening group and jiajia-ji electroacupuncture group(P<0.05).There was no significant difference betwe-en the nerve loosening group and the electroacupuncture group at the same time point(P<0.05).3.The Bcl-xL mRNA and protein expressionThe mRNA and protein expression of Bcl-XL in all groups(except normal control group)decreased continuously with the time prolonging,and reached the lowest value at 4 weeks.At the same time point,the expression of Bcl-XL in the three treatment groups was lower than that in the normal control group(P<0.05),and higher than that in the model control group(P<0.05),and the expression of Bcl-XL in the combination group was higher than that in the jiaji electroacupuncture group and the nerve loosening group(P<0.05).At the same time point,the expression of Bcl-XL in electroacupuncture was higher than that in nerve loosening(P<0.05).4.Bad mRNA and protein expressionDuring 1,2 and 4 weeks,the mRNA and protein expression of BAD in all experimental groups(except the normal control group)decreased continu-ously with the time prolonging,and reached the lowest value at 4 weeks.At the same time point,the expression of BAD in the three treatment groups was higher than that in the normal control group(P<0.05).Lower than model control group(P<0.05);The expression of Bad in the combination group was lower than that in the electroacupuncture group and the nerve loosening group(P<0.05).At the same time point,the expression of BAD was lower than that of the nerve loosening group(P<0.05)Results:Conclusion1.Jiaji electroacupuncture and nerve mobilization can reduce the gastrocnemius muscle cell apoptosis,delay the atrophy of the gastrocnemius muscle in rabbits after sciatic nerve injury,and the combined treatment is better.2.Jiaji electroacupuncture and nerve mobilization can delay gastrocne-mius atrophy in rabbits after sciatic nerve injury.The mechanism may be to inhibit the expression of Bad in gastrocnemius muscle cells and up-regulate the expression of Bcl-xL.
