| ObjectiveThis study looked at how Jiaji electroacupuncture and treadmill exercising affected the expression of IGF-1,AKT,and P-AKT in rats with sciatic nerve transection.These molecules are crucial to the IGF-1/PI3K/AKT signaling pathway.in order to offer the appropriate theoretical foundation,therapy solutions,and strategies for spinal problems based on by peripheral neuropathy.MethodsSixty SD rats were randomly dedicated to one of five groups: normal control,model control,Jiaji electroacupuncture,treadmill training,or Jiaji electroacupuncture+treadmill training(combined group),Each group had 12 rats.The intervention period for the five groups was four weeks.The rat sciatic nerve was completely transected transversely with surgical scissors to establish Sunderland Ⅴ injury model,and then the two sides of the severed end of the adventitia were aligned and sutured with sutures under a microscope.The normal group got no assistance,and the model group got no treatment following detailed modeling.The electroacupuncture group received Jiaji electroacupuncture three days after modeling.The treadmill group received treadmill training three days after modeling.The combined group used the above two treatment methods at the same time.After four weeks of treatment,the behavioral scores of the five groups were measured by the sciatic nerve function index(SFI).After scoring,samples of the five rat groups were taken.The muscle fiber cross-sectional area(FCSA)was measured with HE staining,and Western blot was applied to analyze the concentration of IGF-1,AKT,and P-AKT in the gastrocnemius muscle tissue.Result1.SFIFour weeks following the treatment,the SFI of the rats in the five groups was measured,and the difference was shown to be massively larger in the normal group than in the other four groups(model group,electroacupuncture group,treadmill group,and combination group),with statistically significant differences(P <0.01).When contrast to the therapy groups(electroacupuncture,treadmill,and combination),the model group performed significantly lower(P <0.01).In comparison to the treadmill group and the electroacupuncture group,the combination group performed quite well(P <0.05).Statistically speaking,there was a significant difference between the electroacupuncture and treadmill groups(P <0.01).2.FCSAFour weeks following the treatment,the FCSA was largerin the electroacupuncture,treadmill,and combination groups than in the model group and smaller in the normal group,and the diversity was conclusive(P <0.01).The combination group outnumbered the treadmill and electroacupuncture group by a statistically remarkable margin(P <0.01).The treadmill group outnumbered the electroacupuncture group by a statistically remarkable margin(P <0.05).3.IGF-1 protein expressionFour weeks following the treatment,The proteide communication of IGF-1 in the model and electroacupuncture groups dipped when compared to the normal group,and the diversity was conclusive(P <0.01);the treadmill group was lower than the normal group,but the difference was not markedly significant(P > 0.05);and the combination group was higher than the normal group,with markedly diffeerence(P <0.01).proteide communication elevated in the electroacupuncture,treadmill,and combination groups relative to the model group,with markedly diffeerence(P <0.01).The combination group was better than the treadmill and electroacupuncture groups,with markedly diffeerence(P <0.01).The treadmill group was greater than the electroacupuncture group,with markedly diffeerence(P <0.01).4.Protein expression of AKT and P-AKTFour weeks following the treatment,none of the five groups’ levels of AKT protein expression in the gastrocnemius muscle diverged from one another(P > 0.05).P-AKT proteide communication level: in contrast to the normal group,P-AKT proteide communication was lessened in the model,electroacupuncture,treadmill and combination groups,and the diversity was conclusive(P <0.01).Compared to the model group,proteide communication went up in the electroacupuncture,treadmill,and combination groups;the diversity was markedly significant(P <0.01);among them,the combination group was better than the treadmill and electroacupuncture groups,with markedly diffeerence(P <0.01).The treadmill group was greater than the electroacupuncture group,with markedly diffeerence(P <0.01).Conclusion1.In rats with sciatic neuropathy,Jiaji electroacupuncture and treadmill training may ramp up SFI and the gastrocnemius muscle of FCSA,and avoiding denervated muscle wastage,and treadmill training is preferable than Jiaji electroacupuncture,with a greater combined effect.2.Although Jiaji electroacupuncture and treadmill training can not improve the expression of AKT protein in gastrocnemius muscle,it may delay the atrophy of denervated muscle by ramping up the quantity of IGF-1and P-AKT proteide.Moreover,treadmill training is superior to Jiaji electroacupuncture,and the combined effect of the two is better.3.Delaying denervated muscle atrophy with Jiaji electroacupuncture and treadmill training may be involved with elevated IGF-1 and P-AKT proteide expression. |
