| Objective:By studying the effects of acupuncture-rehabilitation therapy on neurological function and IL-18 and IL-33 protein expression in mice with focal cerebral ischemia,the mechanism of acupuncture-rehabilitation therapy related to brain protection was explored.Methods:According to the random number table method,120 C57BL/6 female mice were divided into five groups: model group,sham operation group,acupuncturet group,rehabilitation group and acupuncture-rehabilitation group.Each group was divided into 24 mice,and each group was divided into3 d and 7d subgroups,namely 12 mice in each subgroup.The permanent cerebral ischemia model was prepared by the modified suture method.The sham operation group and the model group were not given any treatment,the acupuncture group was treated with scalp-point cluster acupuncture,the rehabilitation group received treadmill training,and the acupuncture-rehabilitation group performed treadmill training during the 2hours of scalp cluster puncture and retention of needles.At 3 and 7 days after operation,the degree of neurological deficit in each group of mice was evaluated by the modified neurological deficit score(m NSS),and the changes of serum inflammatory factors IL-18 and IL-33 were detected by enzyme-linked immunosorbent assay(ELISA),and immunohistochemistry and western blot were used to detect the expression of inflammatory factors IL-18 and IL-33 in the ischemic brain tissue of mice.Results:1.m NSS scores of mice in each group: 3 and 7 days after surgery,the sham operation group did not have neurological deficit symptoms,compared with the sham operation group,the m NSS scores of the remaining groups increased significantly(P< 0.05);compared with the model group,the m NSS scores of other treatment groups decreased significantly(P < 0.05);7d after the operation,compared with the acupuncture group and the rehabilitation group,the m NSS score of the acupuncture-rehabilitation group decreased(P< 0.05).2.Serum IL-18 and IL-33 protein content of mice in each group: 3d and7 d after operation,the serum IL-18 content of mice in the sham operation group increased slightly,and the IL-33 content was the highest.Compared with the sham operation group,the levels of IL-18 in the serum of the remaining groups were significantly increased(P < 0.05),and the content of IL-33 was significantly reduced(P < 0.05);compared with the model group,the levels of IL-18 in the serum of other treatment groups were significantly decreased(P< 0.05),and the levels of IL-33 were significantly increased(P< 0.05);compared with the acupuncture group and the rehabilitation group,the serum IL-18 content decreased and IL-33 content increase in the acupuncture-rehabilitation group(P< 0.05).3.The expression of IL-18 and IL-33 protein in the brain tissue of the mice in each group: 3d and 7d after the operation,the basic expression of IL-18 protein in the brain tissue of the sham operation group was the highest,and the expression of IL-33 protein was the highest.Compared with the sham operation group,the expression of IL-18 protein in the brain tissues of the remaining groups increased(P < 0.05),and the expression of IL-33 protein decreased(P < 0.05).Compared with the model group,the expression of IL-18 protein in the brain tissues of other treatment groups was significantly decreased(P < 0.05),and the expression of IL-33 protein was significantly increased(P< 0.05);compared with the acupuncture treatment group and the rehabilitation group,the content of IL-18 in the brain tissue of the acupuncture-rehabilitation group decreased and the content of IL-33increased(P< 0.05).4.The expression of IL-18 and IL-33 protein in the brain tissue of the ischemic penumbra of each group of mice: 3d and 7d after the operation,the basic expression is IL-18 protein and the highest expression is IL-33 protein in the brain tissue of the sham operation group.Compared with the sham operation group,the expression of IL-18 protein in the brain tissues of the remaining groups increased(P < 0.05),and the expression of IL-33 protein decreased(P< 0.05);compared with the model group,the expression of IL-18 protein in the brain tissues of other treatment groups was significantly decreased(P< 0.05),and the expression of IL-33 protein was significantly increased(P < 0.05);compared with the acupuncture group and the rehabilitation group,the content of IL-18 in the brain tissue of the acupunctur-rehabilitation group decreased and the content of IL-33 increased(P< 0.05).Conclusions:1.Acupuncture-rehabilitation therapy can reduce the score of neurological deficit in mice with focal cerebral ischemia,reduce the pathological damage of the ischemic brain tissue and play a protective effect on the brain.2.Acupuncture-rehabilitation therapy can down-regulate the content of IL-18 in brain tissue and serum in the ischemic area of mice with focal cerebral ischemia,and up-regulate the content of IL-33,which can inhibit the inflammatory damage after cerebral ischemia and promote the recovery of inflammation. |
