Objective: The aim of this study is to investigate the effects of Hypericum perforatum on the gastrointestinal kinetics and ENS of FD rats by studying the general status of experimental animals,the relative m RNA expression and protein content of the neuronal marker PGP9.5 in gastric sinus and duodenal tissues,and the specific markers s100β and GFAP in intestinal glial cells,so as to provide new ideas and directions for the treatment of FD.Methods:1.Thirty-six male SD rats of SPF grade were randomly divided into six groups: Group A was the blank group,Group B was the FD model control group,Group C was the FD model administered with Hypericum perforatum high dose group,Group D was the FD model administered with Hypericum perforatum medium dose group,Group E was the FD model administered with Hypericum perforatum low dose group,and Group F was the FD model administered with Mosapride tablets control group.2.The general condition of each group of rats was observed,and body weight was measured once a week.3.The B-F group prepared the experimental animal model by using the compound modeling method of tail-clamping stimulation along with irregular diet.4.After successful modeling,the gastric sinus and duodenum of each model group of rats were taken after two weeks of drug administration for the following tests:Experiment 1: The mean body weight of each group of rats was calculated,and the gastric emptying rate and small intestinal propulsion rate were detected to investigate the effects of Kanto leaf Hypericum on the gastrointestinal kinetics of FD rats.Experiment 2: The m RNA expression of PGP9.5,s100β and GFAP in the gastric sinus and duodenal tissues of rats in each group was detected by real-time fluorescence quantitative PCR(RT-q PCR)method;Gastric sinus and duodenal tissues were homogenized and supernatants were extracted,and the protein content of PGP9.5,s100β and GFAP in the tissues was measured by ELISA.To investigate the effects of Kanto leaf Hypericum perforatum on the ENS of FD rats.Results:1.Gastrointestinal kinetic effects of Hypericum perforatum on FD rats:The weight of rats in each group before modeling was not significantly different(P>0.05);after modeling,the weight of rats in FD model group was significantly lower than that in group A(P<0.05);at the end of drug administration,the weight of rats in each group increased,among which the weight of rats in group D increased most significantly(P<0.05).The gastric emptying rate and small intestine propulsion rate in group B were significantly higher than those in group A(P < 0.05),and they were both higher than those in groups C and F.2.Effect of Hypericum perforatum on the ENS of FD rats:Compared with group A,the expression of PGP9.5 m RNA and protein in the gastric sinus and duodenal tissues of rats in group B was significantly reduced(P < 0.05);compared with group B,the relative expression of PGP9.5 m RNA and protein in the gastric sinus and duodenal tissues of rats in group D was significantly increased(P <0.05).s100β and GFAP m RNA and protein relative expression in the gastric sinus and duodenal tissues of rats in group B were significantly higher than those in group A(P <0.05).The relative expression of s100β and GFAP m RNA and protein in the gastric sinus and duodenum tissues of rats in group B was significantly higher than that in group A(P < 0.05);compared with group B,the relative expression of s100β and GFAP m RNA and protein in the gastric sinus and duodenum tissues of rats in group D was significantly lower(P < 0.05).Conclusions:1.Throughout,Hypericum perforatum increased the body weight of FD rats significantly,promoted their gastric emptying and small intestine propulsion rate,and led to the improvement of the general condition of FD rats.2.Hypericum can increase the relative expression of PGP9.5 m RNA and protein expression,which are abnormally down-regulated,and decrease the relative expression of s100β and GFAP,which are abnormally up-regulated,in the gastric sinus and duodenum of FD rats,thus restoring the damaged ENS of FD rats and improving their gastrointestinal motility disorders.
