| Clinical data of 96 patients with mild bronchial asthma and 100 healthy controls in Mindong Hospital Affiliated to Fujian Medical University were collected and analyzed,and the response of rs242941 gene polymorphism of corticotropin-releasing receptor1(CRHR1)to inhaled corticosteroid treatment was analyzed.Methods:1、By analyzing the general clinical data of 96 patients with bronchial asthma and100 healthy controls in Mindong Hospital Affiliated to Fujian Medical University,the differences were compared.2、The polymorphism of CRHR1 gene rs242941 in the asthma group and the control group was detected by PCR method,and the differences of genotype frequency and allele frequency were analyzed.The gender,age,ACT score,mMRC score,daytime symptom score,nighttime symptom score,diurnal variation rate of PEF,FEV1,FEV1/Pred,FEV1/FVC,PD20-FEV1,FeNO,blood EOS,blood cortisol,blood IL-2,blood total Ig E,positive number of skin allergens,allergy index and other indexes of asthma patients with different genotypes were compared.3、Asthma group was divided according to the results of gene testing,and patients with rs242941 were analyzed and compared before and after treatment for patients with different genotypes,such as ACT score,daytime symptom score,nighttime symptom score,PEF day night variation rate,FEV1 / PRED(%),pd20-FEV1,FeNO,blood EOS,blood IL-2,blood total Ig E,blood cortisol and other indicators.Results:1.In this study,we found that the rs242941 polymorphism of Crhr1 gene existed in patients with bronchial asthma in clinical remission stage and healthy subjects.The genotypes were GG genotype(wild homozygous),GT genotype(heterozygous mutation)and TT genotype(homozygous mutation).The frequency of genotype and allele was not related to gender,age and asthma(P>0.05)It was found that the number of GT and TT genotypes was less,so the dominant model was used to combine GT and TT genotypes.2.There were no significant differences in gender,age,act score,mMRC score,daytime symptom score,nighttime symptom score,PEF day night variation rate,FEV1,FEV1 / PRED,FEV1 / FVC,pd20-FEV1,FeNO,blood EOS,blood cortisol,blood IL-2,Ig E,positive number of skin allergens,allergic index and other indicators between asthma group and control group(P>0.05);3.There were significant differences in act score,daytime symptom score,nighttime symptom score,PEF diurnal variation rate and FEV1 / PRED(%)before and after treatment in asthma group,while there were significant differences in pd20-FEV1 between GG and GT/TT asthma patients with Crhr1 gene rs242941 locus before and after treatment(P<0.05);4.There were no significant differences in FeNO,EOS,IL-2 and Ig E before and after treatment in the asthma group,but there were significant differences in serum cortisol between the two groups before and after treatment.ConclusionIn this study,we found that the rs242941 polymorphism of CRHR1 gene existed in all populations.Inhaled corticosteroids can improve the symptoms of asthma patients,improve the lung function of patients,and reduce the airway hyperresponsiveness,especially in patients with GG asthma at rs242941 site of CRHR1 gene. |
