Background: Asthma is a syndrome of unknown origins that is characterized by abnormal and inflamed mucosa of the airways, wheezing, and shortness of breath. Genetic studies offer a structured means of understanding the causes of asthma as well as identifying targets that can be used to treat the syndrome. Heritability has been estimated as up to 60% and is thought to be determined by genetic factors such as nucleotide variants. People have found a wide variety of related genes have been associated. Interleukin 23, involved in mucosal immunity, is an inflammatory cytokine, which is mainly produced by antigen presenting cells, such as activated macrophages and dendritic cells. IL-23 receptor compound is composed of IL-12 Rbl and new cytokine receptor subunit IL-23 R, and is expressed on a variety of cell surface, such as NK cells, monocytes, MP, DC and T CD4 cells, etc. IL-23 receptor the gene encoding human, located in the region of human chromosome 1 1p31, is associated with asthma, which may affect the response of IL-23. The upregulation of IL-23 receptors by some SNP alleles may enhance the STAT signal transduction and ultimately promote the formation of IL-17, thereby increasing the risk of damage to the immune system.Aim: To investigate the association between single nucleotide polymorphism of interleukin(IL)-23 receptor(IL-23R) gene and children bronchial asthma.Methods: We recruited 70 patients with asthma and 73 controls of the outpatients clinic or inpatients who visited to pneumology department of pediatrics of the first hospital of Jilin University in the period(2014.12-2015.12). We enrolled the research objects who accorded with the inclusion/exclusion criteria. Extract peripheral venous blood 5 samples from the blood samples of the research objects. The genomic DNA was extracted from blood samples by DNA extraction kit, then we configured the connectivity reaction system and amplify PCR. Finally, the 1 microlitre extension product was taken to detect the DNA sequence. Statistical significance was performed using the SPSS19.0 software program.Results:(1) There was no significantly statistical difference in age and sex in two groups between the asthma group and the control group.(2) There were three genotypes of IL-23 R gene rs2201841, namely CC, CT and TT, in asthma group and normal control group. There was no significantly statistical difference in IL-23 R gene rs2201841 locus genotype frequencies in children with asthma and normal control group(χ2=4.383,P>0.05). And the frequency distribution of C and T was significantly different(χ2=4.622,P<0.5).(3) There were three genotypes of IL-23 R gene rs11805303, namely CC, CT and TT, in asthma group and normal control group. There was no significantly statistical difference in both IL-23 R gene rs11805303 locus genotype frequencies and the two alleles frequencies in children with asthma and normal control group(P>0.5).Conclusions:(1) This study found that IL-23 R gene rs2201841 and rs11805303 sites with single nucleotide polymorphisms(SNPs).(2) There was no association between genotypes and asthma, but allele polymorphisms were association with children asthma in Jilin Province.(3) Both the polymorphisms of genotypes and allele were irrelevant with children asthma in Jilin Province.
