| Objective:Meta systematic evaluation of SDC2 gene methylation in the diagnosis of colorectal cancer.Methods:Pub Med,Embase,CNKI,VIP and Wanfang databases were searched.The publication dates were from the establishment of the database to December 31,2020.In strict accordance with the included and exclusion criteria,screening related literature from extraction to extract include the first author,year of publication,country,number of cases in the case group,number of cases in the control group,diagnostic criteria,detection method of SDC2 gene methylation,sensitivity,specificity,calculating the true positive(TP)of each study,false positives(FP),false negatives(FN)and negative(TN)..Quadas-2 was used to evaluate the quality of the included literatures.The QUADAS-2 scale mainly consists of four parts: case selection,trial to be evaluated,gold standard,case flow and progress.Evaluate the risk of migration for all components and evaluate the first three components for clinical suitability.A bivariate meta-analysis model was used to calculate the combined sensitivity,specificity,diagnostic odds ratio(DOR),positive likelihood ratio(PLR),and negative likelihood ratio(NLR),and to plot the composite receiver operating characteristic curve(SROC)curve.Spearman correlation coefficient,Cochran-Q test and I~2 test were used to evaluate study heterogeneity.In addition,sensitivity analysis and subgroup analysis were used to explore potential sources of heterogeneity.Potential publication bias was assessed using Deek’s funnel plot.Results : A total of 178 relevant literatures were retrieved by the two researchers according to the developed retrieval scheme.First,68 literatures were excluded,and 92 literatures unrelated to SDC2 methylation detection in colorectal cancer were screened by reading the title,and the remaining 18 literatures were screened.After reading the abstracts,8 literatures were excluded that could not provide accurate data and the diagnostic criteria were unknown.After reading the full text,10 articles were finally included for Meta-analysis,including 1174 cases in the case group and 1555 cases in the control group.The comorbidity sensitivity was 0.81(95%CI: 0.76-0.86),the comorbidity specificity was 0.94(95%CI: 0.88-0.97),the comorbidity positive likelihood ratio was14.3(95%CI: 6.7-30.8),the comorbidity negative likelihood ratio was 0.2(95%CI:0.15-0.26),the diagnostic odds ratio was 73(95%CI: 31-174),and the area under curve(AUC)of SROC was 0.92.Subgroup analysis showed that SDC2 gene methylation detection in the diagnosis of colorectal cancer,QMSP detection method had higher diagnostic value.Conclusion:Detection of SDC2 gene methylation in feces is of high diagnostic value in colorectal cancer,and it is an ideal marker for early diagnosis of colorectal cancer. |
