| Selenium yeast can be converted into organic active forms represented by selenocysteine and selenomethionine after ingested by the body,which plays an auxiliary anticancer role.The organic selenium content of commercialized yeast selenium is not stable,and the results of studies on its active form participating in the anticancer effect are inconsistent.In this study,Se-rich yeast with high Se-content was obtained by optimizing Se-rich conditions of Saccharomyces cerevisiae from yaks.SEYE-5668 was obtained by snail enzymatic hydrolysis and ultrasonic crushing method,and its Se-content was detected Cell experiment and animal model experiment were used to study the growth inhibition effect of selenium-rich yeast on mouse breast tumors and the regulation effect of selenium-protein m RNA expression in tissue cells.The following results were obtained in this study1.The selenium content of Saccharomyces cerevisiae strain SWUN5668 was increased to 2781μg/g.The breakage rate of yeast cells was increased to 97.62±2.33% by snail enzymolysis combined with ultrasonic crushing.The protein extraction efficiency of no-load yeast and selenium-rich yeast reached 8.70 and 8.35%,respectively(P>0.05),the selenium content of Se-rich yeast extract(SEYE-5668)reached the highest of 3100ng/m L.2.Different types of mouse breast tumor cell lines(4T1,EMT6 and 67NR)showed different sensitivity to different selenium compounds.The inhibitory effect of SEYE 5668 obtained in this study on the three tumor cell lines was significantly stronger than that of other selenium compounds(Sodium Selenite,L-SeC,L-SeM)(P<0.01),and showed the strongest inhibitory effect at 0.01 μmol/L Seconcentration,while the no-load yeast extract had no inhibitory effect in vitro.3.The regulation effect of SEYE-5668 on the m RNA expression of selenoproteome was time-dose-dependent.4.In vivo experiments showed that different selenium source compounds could inhibit the growth of mouse breast tumors(4T1)when overdose was used.Comparison results showed that SEYE-5668 had the most obvious effects on the inhibition of tumor growth and metastasis,and had the least side effects on liver and lung tissues.For the first time in this experiment,it was shown in an in vitro cell experiment that the crushed SEYE-5668 extract has a time-dose-dependent regulation on the expression of western protein group m RNA in breast tumors.In addition,the SEYE-5668 and its broken extracts have shown superior tumor inhibitory effects and smaller side effects than sodium selenite and several other organic substances in in vivo and in vitro tests.SEYE-5668 has broad therapeutic prospects and utilization value in the treatment and prevention of tumor diseases. |
