Objective This experiment aims to evaluate the effect of Ganwei Baihe Decoction on the rat model of chronic stress gastric ulcer(Stagnation of Liver and Stomach Qi)by detecting the behavioral,serological indicators and gastric histopathological changes in rats,and using Metabonomics technology was used to screen the metabolites of gastric tissue in rat models,and to explore the possible mechanism of Ganwei Baihe Decoction in intervention of chronic stress gastric ulcer(Stagnation of Liver and Stomach Qi)in rat models.Methods Thirty-six SPF male SD rats were randomly divided into blank group,model group,and traditional Chinese medicine group(ie Ganwei Lily Decoction group),with 12 rats in each group.The blank group was fed normally.The model group and Chinese medicine group were used to prepare rat models according to the unpredictable chronic stress method.The model was continued for 5 weeks.At the third week of the model,the Chinese medicine group was given traditional Chinese medicine by gavage,and the blank group and the model group were gavage with distilled water.Gave the stomach continuously for 2 weeks.During the experiment,observe the general behavior of the rats,record the weight,water intake and open field test scores,calculate the gastric emptying rate,ulcer index(UI)and ulcer inhibition rate of rats in each group,and observe the pathological changes of gastric mucosal tissue by HE staining.The content of 5-HT in serum of rats was detected by ELISA,and the combined technique of chromatography-mass spectrometry(LC-MS)was used to analyze metabolic markers and metabolic pathways.Results1.After 3 weeks of modeling,the rats in the model group and the Chinese medicine group were in general conditions obviously abnormal,and their weight and scores in the open field experiment were significantly lower than those of the blank group(P<0.05).After gavage,the general condition of the rats in the Chinese medicine group was improved.,Body weight and scores in the open field experiment were higher than those in the model group(P<0.05).2.The gastric emptying rate of rats in the model group was significantly lower than that of the blank group and the traditional Chinese medicine group(P<0.05),and there was no significant difference between the traditional Chinese medicine group and the blank group(P>0.05);the gastric ulcer index of the rats in the traditional Chinese medicine group was significantly lower than that of the model Group(P<0.05),there is no obvious difference from the blank group(P>0.05);the serum 5-HT level in the model group were significantly higher than those in the blank group and the traditional Chinese medicine group(P<0.05).3.It can be seen under the microscope that the gastric mucosa of the model group is significantly congested and swollen,scattered in punctate ulcers,erosions,and more inflammatory cell infiltration than the traditional Chinese medicine group.4.Establish the metabolic spectrum of rat stomach tissue based on the LC-MS combined technology,and analyze the metabolic phenotype changes of rats after the intervention of the liver and stomach qi stagnation syndrome Ganwei Baihe Tang.Use VIP>1 and P<0.05 and FC≥1.2 or FC≤0.8 as the criteria to screen differential metabolites,and then conduct pathway analysis of differential metabolites through Metaboanalyst and KEGG databases and construct a metabolic network.In the administration group VS model group,7 different metabolites were identified in the positive ion mode,and 5 different metabolites were identified in the negative ion mode,including pyrrolidine,uracil,allopurinol,methionine,histidine,and isoleucine Acid,hypoxanthine,xanthine,D-heptaquinone-heptaphosphate,N-acetyl-D-glucosamine-6-phos phate were significantly up-regulated,and Deltaline and adenine were significantly down-regulated.It was found that the liver and stomach lily soup can be Significantly recalled10 potential biomarkers,these potential biomarkers related to amino acid metabolism,energy metabolism and purine metabolism.Conclusion1.Ganwei Baihe Decoction can improve the stress state of model rats,increase the rate of gastric emptying in rats,reduce the levels of 5-HT,promote ulcer healing,and effectively treat chronic stress gastric ulcers(Stagnation of liver and stomach qi))Rats.2.Ganwei Baihe Decoction may promote the repair of rat gastric mucosal tissue and accelerate the healing of ulcers by regulating amino acid metabolism,energy metabolism and purine metabolism,and achieve regulation of inflammation,oxidative stress and HPA axis.The above-mentioned biomarkers are mainly Related to energy metabolism,oxidative stress and inflammation.
