Objective:To establish surface activation molecules of T lymphocytes in peripheral blood of healthy adults in Liaoning province:CD4~+CD38~+,CD8~+CD38~+,CD4~+HLA-DR~+,CD8~+HLA-DR~+,differentiation and memory molecules:CD4~+CD28~+,CD4~+CD28~-CD57~+,CD8~+CD28~+,CD8~+CD28~-CD57~+,CD4~+CD45RA~-RO~+,CD4~+CD45RA~+RO~-,CD8~+CD45RA~-RO~+,CD8~+CD45RA~+RO~-;depletion related molecules:CD4~+PD-1~+,CD8~+PD-1~+to analyze whether there are differences between men and women,the trend of peripheral blood T lymphocyte related indicators with age,and Analyze whether there is any difference between the results obtained by the two mainstream instrument platforms under the same sample,the same operation,and the same experimenter,and determine whether these indicators need to establish separate reference intervals according to gender,age,and instrument,provide reference for the research of immune system function,diagnosis and prognosis of various diseases.Methods:70 healthy adult people based on normal whole blood count and liver and kidney function tests.Flow cytometry was used to detect CD4~+,CD8~+T cell surface activation molecules HLA-DR,CD38,differentiation and memory related molecules CD45RA,CD45RO,CD28,CD57,depletion-related molecule PD-1 and regulatory T cell expression levels;and divide the population into groups according to age(youth group:≤30 years old,middle-aged group:31 to 49 years old,elderly group:≥50 years old).Analyze the differences between the above indicators in different ages,different genders and different instrument platforms.Results:1)Seventy eligible healthy adults were selected,ranging in age from 24 to 86years old.Among them,there were 10 cases in the youth group(24 to 30 years old),with an average age±standard deviation of 28±2 years,and 34 cases in the middle-aged group(31 to 49 years old),with an average age±standard deviation of 41±6 years.26 cases in the elder-aged group(50 to 86 years old),the average age±standard deviation is 62±11 years;There were 35 males and females,establish the above 14 research indicators Reference interval according to the small sample(<120cases)robust method from the C28A3 standard document;2)If the results measured on the two platforms conform to the normal distribution,the independent speciment test is used.Otherwise,the Mann-Whitney test is used.CD4~+PD-1~+,CD8~+CD38~+measured on the instrument BD FACSCantoⅡhave statistically significant differences between genders,the other results are not statistically significant between genders(p>0.05);The differences between gender of CD8~+CD28~-CD57~+,CD8~+CD38~+measured on the instrument Beckman Navios are statistically significant.The remaining results are not statistically different between genders(p>0.05).3)A one-way ANOVA was used between the three age groups.In the analysis of the two platforms,the ratio of CD8~+CD38~+decreases with age;The ratio of CD4~+CD38~+in the elderly group is slightly lower than that in the middle-aged group;the ratio of CD4~+HLA-DR~+increases with age;there is no difference in other indicators among the groups4)If the results measured on the two platforms conform to the normal distribution,the independent specimen t test is used.Otherwise,the Mann-Whitney test is used.Treg,CD4~+PD-1~+,CD8~+PD-1~+,CD4~+CD45RA~-RO~+,CD4~+CD45RA~+RO~-,CD4~+CD38~+have statistically significant differences between the two platforms,and there was no statistically significant difference in other indicators between the two mainstream testing platforms.Conclusion:The reference interval for CD4~+PD-1~+,CD8~+PD-1~+was established for the first time,and the reference interval of peripheral blood T lymphocytes CD4~+PD-1~+、CD8~+PD-1~+、CD4~+CD28~+、CD4~+CD28~-CD57~+、CD8~+CD28~+、CD8~+CD28~-CD57~+、CD4~+CD45RA~-RO~+、CD4~+CD45RA~+RO~-、CD8~+CD45RA~-RO~+、CD8~+CD45RA~+RO~-、CD4~+CD38~+、CD8~+CD38~+、CD4~+HLA~-DR~+、CD8~+HLA~-DR~+used in clinically used in Liaoning and other healthy adults with similar demographic characteristics was established for the first time.the T cell related indicators measured on the BD FACSCantoⅡand Beckman Navios platforms were compared for the first time.
