Objectives Long non-coding RNAs(lnc RNAs)can inhibit the negative regulation of m RNAs through competitive binding of miRNAs.Such interactions between lnc RNAsmiRNAs-m RNAs constitute a network of competing endogenous RNAs(ce RNAs),and are involved in tumor genesis and development.In recent years,many studies have shown that lnc RNAs play an important role in the development of papillary thyroid cancer(PTC).This study aims to analyze the expression of lnc RNA CASC2 in PTC and its correlation with the clinicopathological characteristics of PTC,and to construct its ce RNA regulatory network by using bioinformatics analysis and RT-PCR methods,so as to explore the possible molecular mechanism of lnc RNA CASC2 in the occurrence and development of PTC.Methods 1 The research collected separately 50 cases of cancerous tissues,confirmed as PTC by pathological tests,and adjacent tissues,from January 2020 to October 2020,and used RT-PCR to detect the expression of lnc RNA CASC2.Analyzing the correlation between lnc RNA CASC2 and clinical features of PTC was of primary importance.2 The targeted miRNAs of lnc RNA CASC2 and target genes of candidate miRNAs were obtained through starbase bioinformatics website,and the expression correlation was screened using Pan-Cancer transcriptome sequencing data after TCGA database integration,and the miRNAs and m RNAs with the highest correlation coefficient were selected.3 We struggled to make further detection the expression of miR-155-5p and FOXO1 in local clinical samples,and analyze the relationship between expression and correlation in PTC.Results 1 The expression of lnc RNA CASC2 in PTC tissues is significantly lower than that in normal tissues adjacent to cancer [median1.22(0.893)vs2.021(0.656),P<0.05],which is significantly negatively correlated with tumor size,lymph node metastasis,and clinical staging and irrelevant to age and gender.2 We screened the lnc RNA CASC2 target gene miR-155-5p through the starbase bioinformatics website,and the correlation coefficient between the two was-0.432 by Pan-Cancer analysis;We used the starbase bioinformatics website to screen the miR-155-5p target gene FOXO1,and the correlation coefficient between miR-155-5p and FOXO1 was-0.239,and that between lnc RNA CASC2 and FOXO1 was 0.531.3 The expression of miR-155-5p in PTC tissues was equally higher than that in normal tissues adjacent to cancer [median4.931(2.005)vs 1.551(1.318),P<0.05].The expression of lnc RNA CASC2 and miR-155-5p was significantly negatively correlated(r=-0.529,P<0.01);the expression of FOXO1 in PTC tissues was significantly lower than that in normal tissues adjacent to cancer [median1.653(0.840)vs 2.892(1.713),P<0.05].lnc RNA CASC2 was positively correlated with FOXO1 expression(r=0.485,P<0.01),and miR-155-5p was significantly negatively correlated with FOXO1expression(r=-0.533,P<0.01).Conclusions 1 The expression of lnc RNA CASC2 in PTC is significantly downregulated,promoting the occurrence and development of PTC and presenting the significantly negative correlation with tumor size,lymph node metastasis,and clinical stage.2 Biological information analysis indicated that miR-155-5p has binding sites of lnc RNA CASC2 and FOXO1,and the expression of lnc RNA CASC2 and FOXO1 are positively correlated,whereas are negatively correlated with the expression of miR-155-5p.All of our test results substantiate the suspect that lnc RNA CASC2 may be involved in the occurrence and development of PTC by means of miR-155-5p-FOXO1 regulatory network.Figure8;Table30;Reference 152...
